Objective:To determine whether Jian pi li qi decoction is effective for the apoptosis rate of stomach smooth muscle cells and genes bcl-2/bax expressionfor diabetic gastroparesis rats(DGP).Method:The DGP model was established.Wistar rats were divided into five groups:the normal group and the model group were treated with saline,the controlled group with Motilium suspension,the two treatment groups with high or low dose of Jian pi li qi decoction respectively.After three-week continuous intragastric administration,the following outcomes were measured after treatment:body weight,blood glucose,concentration of gastric dynamic element in plasma,the rate of gastric emptying,apoptosis rate of gastric tissue cells,genes bcl-2/bax protein expression.Results:The weight of DGP rats were significantly lower than that of the normal group(P<0.05),Jian pi li qi decoction and Motilium both increased the body weight of DGP rats.The average blood glucose of DGP rats were significantly higher than those of the normal group(P<0.001),Jian pi li qi decoction reduced the blood glucose compared to the model group(P<0.05),the blood glucose of high-dose Jian pi li qi decoction group was higher than that of other DGP rats(P<0.05).The rate of gastric residue detection and motilin concentration in plasma of the high-dose Jian pi li qi decoction and normal group were all lower compared to the model group.The high-dose Jianpi liqi group was lower than the low-dose group and Motilium controlled group(P<0.05),but had no significant difference compared with the normal group(P>0.05).The smooth muscle cells in gastric tissue of DGP rats were showed disorder arrangement and vacuolar degeneration.The above pathological changes in model group and Motilin group were the worst,that of the low-dose group was in the second place,that of the high-dose group was light.Gastric smooth muscle cells apoptosis in DGP rats were significantly increased compared to the normal group(P<0.001),that of the model group was significantly higher than other DGP groups(P<0.05),but no significant difference was found compared with Motilin group(P>0.05),the apoptosis rate in high-dose group was lower than that of the other DGP rats(P<0.05).The results of protein expression of genes bcl-2 and bax in gastric smooth muscle cells showed that the protein expression of these two genes was negative correlation.The protein expression of bcl-2 and bax showed obvious abnormal in the model group.Compared to the low-dose Jian pi li qi decoction group and the controlled group,bax gene expression was significantly reduced(P<0.05),and the bcl-2 gene expression was obviously increased in the high-dose group(P<0.05).Conclusion:Compared to normal rats,DGP rats showed lower body weight,higher blood sugar and obvious gastric motility disorder.High-dose Jian pi li qi decoction showed good effects on increasing the body weight,reducing blood sugar and residual rate of gastric pigment,improving the secretion of motilinin plasma and the pathological changes of gastric smooth muscle cells for DGP rats.The protein expression of Bcl-2 and Bax in gastric smooth muscle cells in DGP rats showed obvious abnormal and was negative correlation.Jian pi li qi decoction can benign regulate genes Bcl-2/Bax expression for diabetic gastroparesis rats,which may be one of the effective mechanism.
