| Objective:In this study,we detect the STAT3,NF-kBmRNAin peripheral blood CD4+T and STAT3,NF-kB protein in the plasma expression levelsbetween Xinjiang Uygur,Han population with asthma and healthy population, compared the correlation of the gene expression levels to explore the role of STAT3 and NF-kB in the occurrence of asthma.Methods:(1)We recruited the study subjects diagnosed with acute attacked of asthma from the First Affiliated Hospital of Xinjiang Medical University, which included 157 patients(asthma group).In the same period, 121 healthy people who had the physical examination in the First Affiliated Hospital of Xinjiang Medical University were also selected (control group).(2)We applied the real-time quantitative RT-PCR to detect the STAT3 and NF-kBmRNA expression levels.(3)We also appliedthe enzyme-linked immunosorbentto detect the STAT3 and NF-kB protein expression levels.Results:(1)In asthma group,the expression levels of STAT3mRNA and protein were higher than the control group,and there had statistically significant difference(P<0.05). In wind phlegm asthma group,cold asthma group and heat asthma group,the expression levels of STAT3mRNA and protein were higher than the control group, and there had statistically significant difference(P<0.05),but the expression levels of STAT3mRNA had no statistically significant difference between the wind phlegm asthma group,cold asthma group and heat asthma group(P>0.05).Between the Uygur asthma group and Han asthma group, the STAT3mRNA expression had significant difference(P<0.05),but the STAT3 protein expression had no significant difference(P>0.05).(2)In asthma group,the expression levels of NF-kBmRNA and protein were also higher than the control group,and there had statistically significant difference(P<0.05).In wind phlegm asthma group,cold asthma group and heat asthma group,the expression levels of NF-kBmRNA and protein were higher than the control group,and there had statistically significant difference(P<0.05),but the expression levels of NF-kBmRNA had no statistically significant difference between the wind phlegm asthma group,cold asthma group and heat asthma group(P>0.05).Betweenthe Uygur asthma group and Han asthma group,the NF-kBmRNA and protein expression had no significant difference(P>0.05).Conclusion:(1)STAT3 and NF-kB are involved in the pathogenesis of asthma.(2)STAT3 may be the reason of the different morbiditybetween Uygur asthma and Han asthma.(3)Our study showed that there were no obvious relationship between the wind phlegm asthma,cold asthma,heat asthma and STAT3, NF-kB. |
