Purpose:To explore the function of Invigprating the spleen to Benefit Qi prescriptions prescriptions on asthma model with spleen-deficiency, via observing IL-12, IL-13 levels and the expression of IKKα. Study the relationship amony IL-12, IL-13 levels, expression of IKKα, pulmonary function, in order to further reveal the mechanism of its anti-asthmatic function and clinics significance .Material and method:40 healthy male Wistar rats, body weight 200±20g, months of age 6-8 weeks, were randomly assigned into 5 groups, n = 8,there are:normal group,spleen deficient asthmatic model group, asthmatic model group,spleen deficient asthmatic model treated by Traditional Chinese Medicine group and asthmatic model treated by Traditional Chinese Medicine。First, using improper diet,over-fatigue method and rhubarb on spleen deficient asthmatic model group and spleen deficient asthmatic model treated by TCM group to establishment spleen deficient model for 10 days. Then, with asthmatic model group and asthmatic model treated by TCM group were ovalbumin sensitized and-challenged way to establish the asthmatic model. 11 days later, spleen deficient model were build, so that, started to i.g Bupiyiqi Fang to spleen deficient asthmatic model treated by TCM group and asthmatic model treated by TCM for 21 days.The way of HE staining was used to observe the condition of EOS; Animal lung ventilator and pulmonary function analysis software were applied on measuring the respiratory function of lung;The levels of IL-12,IL-13 in BALF were detected by the method of enzyme-linked immunosorbent assay (ELISA); the expression of IKKαin lung tissue and airway was observed by immunohisto chemistry technique.Results:Airway responsieness, at 80ug/kg of ACH quantity, spleen deficient asthmatic model group and asthmatic model group, Re doubled obviously (Re>2), the Re of two treatment groups and normal group had no similar reactions(P<0.01). At 160u g/kg of ACH quantity, spleen deficient asthmatic model group, asthmatic model group, spleen deficient asthmatic model treated by Traditional Chinese Medicine group, asthmatic model treated by Traditional Chinese Medicine, Re doubled obviously, spleen deficient asthmatic model group was the highest value, two treatment groups were the lowest value(P<0.01), the Re of normal group had no similar reactions (P<0.01); HE staining, trachea and alveolar structures of normal group were normal, around bronchus of lung tissue were plenty of eosinophilic infiltrate, bronchial mucosal folds, mucosal surfaces, thickening airway narrows, alveolar cavity secretion increased significantly, asthmatic model group is slightly lighter than spleen deficient asthmatic model group, these symptoms of two treatment groups were significantly reduced. Immunohistochemical and ELISA results, compared with the normal group, the expression of IKKα,IL-13 of asthmatic model group and Spleen deficient asthmatic group were significantly higher (P<0.01) in statistics, however, IL-12 was lower (P<0.01). The IKKαexpression and IL-13 levels of Spleen deficient asthmatic group were higher than the asthmatic group (P<0.05). While those two groups treated by TCM's were significantly lower than spleen deficient asthmatic and asthmatic model group (P<0.01). There was a significant statistical difference, IL-13 were significantly higher (P<0.01), nevertheless, IL-12 was statistically higher there than. Yet, comparing the two TCM treated groups, IL-13 and the expression of IKKαof asthmatic group treated by TCM were lower than Spleen deficient asthmatic treated by TCM, in contraries, IL-12 levels was higher.Conclusion:1. Spleen deficient can increase asthma.2. Invigprating the spleen to Benefit Qi prescriptions can reduce HE dyeing and airway responsieness index on asthma model with spleen-deficiency rat.3. Invigprating the spleen to Benefit Qi prescriptions can make the IKKαof lung and bronchus tissues down-regulated, and regulat NF-κB transcription factors, make the gene transcription factor of inflammation by certain inhibition.4. Invigprating the spleen to Benefit Qi prescriptions can raise IL-12, cut IL-13, reverse IL- 12/ IL-13 ratio imbalance.
