Baicalin isolated from traditional Herba skullcap was used as the research object to explore the regulation of the imbalance of Th17/Treg effects in allergic asthma mice.The mouse were divided into blank group, positive control group, baicalin group (low, medium, and high doges), model group. OVA and LPS were used to sensitive to build model of asthma based on Th17 immune response, by respiratory tract.24 h after the last sensitization, mice were sacrificed, lungs were weighed and organ coefficient was calculated. The pathological changes and mucus secretion of lung tissue were observed in mice. The concentrations of anti-OVA specific IgE in serum, IL-17A, IL-6 and IL-10 in BALF of mice were measured by ELISA. Immunohistochemistry and western blot method were used to detecte the expression of STAT3 and FOXP3 protein in lung tissue of mice.The result showed:Compared with model group, baicalin in medium and high dose groups significantly reduced mucus secretion, decreased organ coefficient of lung tissue and the inflammatory responses, which showed significantly different (P < 0.01 or P< 0.05), but baicalin in low dose group decreased organ coefficient of lung tissue was not significant (P>0.05).Compared with model group, baicalin in low, medium and high dose groups could significantly inhibit the secretion of anti-OVA specific IgE in serum of asthmatic mice, which showed significantly different (P< 0.01). Baicalin in low, medium and high dose groups could significantly inhibit the secretion of IL-17A in BALF of asthmatic mice, which showed significantly different (P<0.01 or P<0.05). Baicalin in medium and high dose groups could significantly inhibit the secretion of IL-6 in BALF of asthmatic mice, which showed significantly different (P<0.01), but baicalin in low dose group was not significant (P>0.05). Baicalin in high dose group could significantly promote the secretion of IL-10 in BALF of asthmatic mice, but baicalin in low and medium dose group was not significant (P>0.05).Compared with model group, baicalin in medium and high dose groups could significantly inhibit the expression of STAT3 protein in the lung tissue of mice, which showed significantly different (P<0.01 or P<0.05), but baicalin in low dose group was not significant (P> 0.05). Baicalin in low, medium and high dose groups could significantly promote the expression of FOXP3 protein in the lung tissue of mice, which showed significantly different (P<0.01 or P<0.05).Conclusion:Baicalin could inhibit the secretion of anti-OVA specific IgE on the imbalance of Th17/Treg response in serum of asthmatic mice, inhibit the secretion of IL-17A and IL-6 in BALF of asthmatic mice, reduce inflammation of lung tissue, reduce mucus secretion, inhibit the expression of STAT3 proteins and promote the expression of FOXP3 protein in the lung tissue of mice.
