Part I Animal model of traumatic myositis ossificans of quadriceps femorisObjective To establish an animal model of traumatic myositis ossificans by repeatedly striking the quadriceps and fixing the knee joint.Methods 18 Adult white New Zealand rabbits,the left lower limb was used as the model group and the right lower limb as the control group.Model group in the 100 cm height with 0.25 kg steel balls hit the middle of the left lower quadriceps,resulting in left lower limb quadriceps soft tissue injury,hit once every 3 days,after 4 weeks to stop the hit,extension of fixed left knee.In the control group,only the right knee joint was fixed.Regular observation of local signs,muscle swelling,local ossification.The animals were sacrificed at 4,6,and 8 weeks(n=6).The pathological changes of quadriceps muscle were observed and analyzed statistically.Results In model group,the swelling rate of the quadriceps was significantly higher than control group(P <0.05),the knee joint was stiff,stiff and localized.The control group had stiff joint,But the quadriceps did not touch the local hardening.The Imaging examination showed that at the 4th week,the model group began to appear the blurred cloudy high-density shadow,the control group did not appear;Week 6 and 8 weeks,with the increase of time,ossification lesions increased gradually,there was significant ossification shadow in model group,the control group no ossification lesions appear.In model group,HE staining showed that osteoblasts and bone tissue formation in the muscle tissue of the quadriceps muscle at the 4th week.There was obvious cartilage or bone formation at the 6th week,and the mature bone tissue was formed at the 8th week.Conclusion Repeatedly hitting the quadriceps and fixing the knee can successfully establish the animal model of traumatic myositis ossificans after pure muscle injury.Part II Study of Chinese herbal medicine on the prevention of traumatic myositis ossificans after muscle repeat injuryObjective To investigate the preventive and therapeutic effects of traditional Chinese medicine on traumatic ossifying myositis(TMO)induced by repeated muscle injury.Methods Eighteen adult New Zealand white rabbits were divided into treatment group and control group,the lower limbs of the same body were the treatment group and the right lower limb was the control group.With a weight of0.25 kg iron ball in the height of 100 cm vertical hit the middle of lower limb quadriceps 3 times,every 3 days hit once,causing the quadriceps muscle repeated injury,straight Fixed knee knees.The treatment group(left lower limb)application of traditional Chinese medicine after each hit,the control group(right lower limb)without any treatment after 4 weeks to stop the hit.Regularly observe the degree of swelling,local signs,local ossification.Animals(n=6)were sacrificed at 2,4and8 weeks,respectively,and imaging,pathology,histology and statistical analysis were performed.Results The swelling rate of the quadriceps muscle of the control group was significantly higher than that of the treatment group(P <0.05).The muscle of the control group could reach the muscle lumps in the control group;the treatment group was not touched;CT three-dimensional imaging showed that at the same time,the control group had obvious ossification,while the treatment group did not appear bone shadow.HE staining showed that the control group was hit by muscle at the bone,cartilage formation,significant ossification;treatment group was not found osteogenesis.Conclusion External application of traditional Chinese medicine can effectively prevent traumatic ossifying myositis(TMO)caused by muscle injury.
