Objective:Using the Achilles tendon induced traumatic myositis ossificans building method, to establish animal model of SD rats by clinical commonly used specific as a treatment drug, Qizaoruanjianruangao by rat Achilles tendon local topical drug intervention therapy, to explore Qizaoruanjianruangao in ossification degree of influence, on the one hand, further verify the Qizaoruanjianruangao effect on prevention and cure of traumatic myositis ossificans, provide theoretical reference for clinical medication. The other side try to investigate the formation mechanism of traumatic myositis ossificans, and also makes a preliminary explore the mechanism of curative effect on the drug, provide theoretical basis for clinical prevention and treatment of myositis ossificans.Methods:This study selected 84 male SD rats, rats all line on the right side of the Achilles tendon amputation, myositis ossificans animal models induced by trauma, postoperative chosen according to random number table method 80 rats were randomly divided into experimental medicine group and blank control group B, each group 40 (n= 40), only the remaining 4 rats as A spare. Experimental group A before 3 weeks do not make any processing, conventional breeding, starting from 4 weeks to local topical drug intervention, chitin,1 day,1 day off per week, apply 7 hours at A time, to intervene to 10th at the weekend. Blank control group B don’t make any deal with the whole experiment process, building back after feeding cage conventional breeding. Two groups in postoperative 3,5,7,9,10 weeks at the weekend, a week to each eight (8*5*2=80) line of X-ray, heart blood of rats in the rat serum Ca, P content, and then in the Achilles tendon, comparative observation of postoperative and normal rat Achilles tendon forms (color, size, flexibility, softness and ossification location), and fixed with 10% formaldehyde solution, timely send paraffin section, HE stain in light downward after morphological observation, histological evaluation of the expression of BMP2 and immune level. Using SPSS 21.0 software to deal with the data from experiment.Results:After the third weekend of 1, building all rats X-ray no bone formation and density increase, two groups of rats in 9 ossification all appear when at the weekend. In building 3,5,7,9, 10 (5 phase point) at the weekend grading score line X-ray results, two groups of ossification of the Ridit analysis, drug group in 5,7,9,10 weekend were higher than blank control group (P<0.05).2, respectively after the two groups in five phase points removed surgically rat Achilles tendon tissue morphology observation,5,7,9, 10 weekend compared the two groups, treatment group A tendon darker, more bulky volume, flexibility and elasticity is relatively poor; 3, two groups of rats serum calcium, phosphorus content in each phase respectively lines between groups, and two more in the group had no significant difference (P> 0.05), but observation of calcium phosphate content of lifting dynamic changes in the whole experiment process, two groups of rats serum calcium phosphate were falling from the fifth weekends, fell to a weekend for minimum 7, then gradually increases, basically the same change trend; 4, histological examination in the two groups were not seen in the third weekend ossification, but have A lot of inflammatory cells and connective tissue hyperplasia, weekend 5 drugs compared with the control group B, group A higher incidence of cartilage and osteoblast,7 at the weekend, both two groups of chondrocytes and osteoblasts, A osteoblast more medication group,9 weekend, treatment group than the control group B in early mature bone matrix and bone trabecular,10 weekends treatment group A, bone tissue more mature.5, histological half quantitative analysis to evaluate the immune, medication group A in A more positive expression of BMP2 positive cells number and staining intensity were higher than control group B.Conclusion:1, To cut off the rat Achilles tendon induced traumatic myositis ossificans effectively, this building method, though with a long cycle experiment, but simple, experiment system control and good repeatability, stable and reliable results.2, Serum calcium, phosphorus content change and the rat Achilles tendon closely related to the formation of traumatic myositis ossificans; 3, BMP2 in rats with traumatic myositis ossificans Achilles tendon has played a role in the formation process of; 4, Qizaoruanjianruangao to lighten the ossification, aggravate the ossification degree may be associated with excessive force in the process of the improper operation.
