Objective:Detected the K-ras and Survivin protein expression in colorectal cancer using immunohistochemical method,and K-ras gene mutation was detected by direct sequencing.Then sorting clinical data and marching 5 years of follow-up after the operation.Preliminary discussion on the relationship of survivin protein and K-ras protein,s expression and mutation in colorectal cancer.Preliminary discussion on the relationship between the two genes and colorectal cancer pathological parameters and its influence on prognosis.Methods:We selected 53 cases of radical resection of colorectal cancer in the First Hospital of Jilin University from March 2010 to November 2011.After 5 years of follow-up.The experimental group 53 cases is the carcinoma tissue specimens.The control group 53 cases is the paracancer tissue specimens far away from the carcinoma tissue 5 cm(the postoperative pathology confirmed is normal tissue).Selected the tissue samples of wax block from pathology department and tissue specimen bank.We cleared up the materials of experimental clinical cases according to age,T stage,histological type,sex,vascular invasion,lymph node metastasis number clinical in experimental group.Detection of tissue samples waxblock slice the expression of suvivinand K-ras protein using immunohistochemical method and K-ras gene mutation was detected by direct sequencing.We used the statistics software SPSS 22.0 to analyze the data.Categorical data analysis using chi-square test,spearman correlation analysis for correlation analysis,Kaplan-Meier methods for survival analysis,?= 0.05 as the inspection standard and P<0.05 difference was statistically significant.Results:1.The expression of survivin protein in carcinoma tissue was significantly higher than that in the adjacent normal tissues(P < 0.05)and its protein expression was related to histological type,lymph node metastasis and T staging(P < 0.05),was not related to the age,gender,vascular invasionand other clinical and pathological parameters.2.In carcinoma tissue the expression of K-ras protein was significantly higher than that in the adjacent normal tissues(P < 0.05)and its protein expression was related to vascular invasionand,lymph node metastasis(P < 0.05),was not related to the age,histological type,gender,T staging other clinical and pathological parameters.3.The mutation rate of K-ras gene was 39.62%,and it was related to lymph node metastasis(P < 0.05),was not related to the age,T staging,histological type,gender,vascular invasionand and pathological parameters.4.Survivin protein,K-ras protein expression in the tissue of colorectal cancer has no obvious correlation(r = 0.223,P > 0.05)?The expression of Survivin protein was correlated with K-ras gene mutation(?2= 9.877,P < 0.05).5.In the 5 year survival rate of colorectal cancer patients,the survival rate of K-ras and Survivin protein positive expression group was significantly lower than that of negative expression group,and the difference was significant(P<0.05).Conclusions:1.The expression of Survivin and K-ras protein in colorectal cancer and normal tissues were different.2.The expression of Survivin and K-ras genes may be independently of each other affects the development process of colorectal cancer,which needs further study.3.The high expression of Survivin gene is associated with the mutation of K-ras gene,which can promote the development,invasion and metastasis of colorectal cancer.4.The expression of Survivin and K-ras protein changes associated with the prognosis of colorectal cancer radical surgery.
