Objective: This essay aims to find out the occurrence, development and invasion,metastasis and its clinical significance in colorectal cancer by the study of detection ofthe expression and its correlation of TCF-4, MMP-7and Survivin in colorectal cancer.Methods:1.Application of reverse transcription polymerase chain reaction (RT-PCR) technique in50cases of colorectal cancer and its corresponding adjacent normal mucosa was to testthe expression mRNA level of TCF-4, MMP-7and Survivin, and analyze therelationship between the expression mRNA level of TCF-4and clinical pathologicalcharacteristics of colorectal cancer.2.The immunohistochemical staining technique (Elivision) was used in100cases ofcolorectal cancer and60cases of normal mucosa tissues to detect the expression proteinlevel of TCF-4, MMP-7and Survivin, and to analyze their relationship with clinicalpathological characteristics of colorectal cancer.Results:1.RT-PCR: The expression mRNA level of TCF-4was respectively0.6527±0.1895and0.2173±0.0452in50cases of colorectal cancer tissues and their normal mucosa tissues,this difference was statistically significant (P<0.01); the expression mRNA level ofMMP-7was respectively0.8236±0.2183and0.1682±0.0384, this difference wasstatistically significant (P<0.01); the expression mRNA level of Survivin wasrespectively0.5418±0.1951and0.1845±0.0639, this difference was statisticallysignificant (P<0.01); the expression mRNA level of TCF-4in colorectal cancer wasrelated to Dukes stages of patients (P<0.05).2.Immunohistochemical Method: In100cases of colorectal carcinoma tissue and60cases of adjacent normal mucosa tissue, the expression protein level of TCF-4incolorectal cancer tissues was higher than that in normal colorectal mucosa tissues. Andits positive expression rate was respectively69%(69/100) and16.67%(10/60), and thedifference was statistically significant between the two groups (P<0.01). The expressionprotein level of TCF-4in colorectal cancer was related to lymph node metastasis andDukes stage of patients (P<0.05). The expression protein level of MMP-7in colorectalcancer tissues was significantly higher than that in normal colorectal mucosa tissues. And its positive expression rate was respectively77%(77/100) and13.33%(8/60), andthe difference was statistically significant between the two groups (P<0.01). Theexpression protein level of MMP-7was related to the depth of invasion,lymph nodemetastasis and Dukes stage of patients(P<0.05). The expression protein level ofSurvivin in colorectal cancer tissues was higher than that in normal colorectal mucosatissues. And its positive expression rate was respectively65.00%(65/100) and15.00%(9/60), and the difference was statistically significant between the two groups (P<0.01).The expression protein level of Survivin was related to lymph node metastasis andDukes stage of patients (P<0.05).3.The correlation analysis of Spearman shows that in colorectal cancer tissues, thepositive expression protein level of TCF-4and MMP-7was positive correlation(r=0.302,P<0.01), TCF-4and Survivin was positive correlation (r=0.279,P<0.01),MMP-7was not associated with Survivin (r=0.097,P>0.05).Conclusion:1.The expression level of TCF-4, MMP-7and Survivin were all higher than normalmucosa tissues, it suggested that they may be involved in the occurrence anddevelopment of colorectal cancer.2.The expression level of TCF-4, MMP-7and Survivin were all related to lymph nodemetastasis and Dukes stages of patients with colorectal cancer, the expression proteinlevel of MMP-7was also related to the depth of invasion. It suggested the three may beinvolved in the malignant growth and clinical progression of colorectal cancer.3.In colorectal cancer tissues, the expression protein level of TCF-4and MMP-7,Survivin was positive correlation respectively, it suggested that the activation of Wntsignaling pathway of nuclear transcription factor TCF-4may play a promoting effect onthe abnormal activation of downstream target gene MMP-7and Survivin. Theexpression protein level of MMP-7was not associated with that of Survivin, itsuggested that they may both be the independent risky factors of the occurrence anddevelopment of colorectal cancer.4.TCF-4, MMP-7and Survivin not only are expected to become the diagnosis andprognostic markers for colorectal cancer, but also provide a new method for targetedtherapy of colorectal cancer.
