| Objective:Examine the relationship between Beclinl, survivin and the. generation, development, invasion and transfer of colon cancer by detecting of the expression of the autophagy genes-Beclin1and Survivin-in the colorectal cancer tissue and by analyzing the clinical pathology data.Method:Choose50paraffin blocks of colorectal cancer tissue which is gotten from surgery and was confirmed by pathology. Choose another30paraffin blocks of normal large intestine mucosal tissue which is5cm away from the cancer tissue. All of these paraffin blocks come from the patients of affiliated hospital of Yanbian. medical University from Jan2010to Jan2014and they all have complete clinical data "and pathological files. The patients of the colorectal cancer tissue samples didn’t get any radiotherapy chemotherapy treatment before surgery. Analyze the correlation between indicators of colorectal cancer tissue’s pathological pattern, Infiltration degree, lymphatic metastasis, ect and the expression of Beclin1and survivin through streptomycete Immunohistochemical SP method.Results:(1) The positive expression rate of Beclin1in the Colorectal cancer tissue is38%, and this is significant lower than the Beclin1positive expression rate (73%) in the normal large intestine mucosa tissue beside the colorectal cancer tissue. The expression of Beclinl is not related to gender, age, tumor site and the depth of invasion (p>0.05), but it is connected with the differentiated degree and lymphatic metastasis of the Colorectal cancer tissue (p<0.05).(2) The Positive expression rate of Survivin in the Colorectal cancer tissue is66%which is obvious higher than the positive expression rate (3%) in the normal large intestine mucosa tissue beside the colorectal cancer tissue (p<0.05). The expression of Survivin is not related to gender, age, tumor site and the depth of invasion (p>0.05), but it is connected with the differentiated degree and lymphatic metastasis of the Colorectal cancer tissue (p<0.05).(3) The expression of Beclin1and Survivin in the Colorectal cancer tissue is correlative (R=-0.416P=0.013).Conclusion:(1) The expression of Beclinl in the colorectal cancer tissue is obvious low and the expression of Survivin in the colorectal cancer tissue is significant high, which indicates that the expression of Beclinl and Survivin may impact the generation, development, invasion and transfer process of colorectal cancer.(2) Beclin1and Survivin abnormal expression in the colorectal cancer may promote the occurrence and development.(3) Joint detection of protein expression level of the Beclinl and Survivin could be used to evaluate the prognosis of patients and provide the basis for clinical treatment. The Beclin1and Survivin may represent new molecular targets for therapy of the colorectal cancer. |
