ObjectiveLooking for the processing technology' s crucial factors by testing the key processes of Leonurus japonicus' s processing technology and uniform design methodents, normalize the processing technology of Leonurus japonicus and enhance the products' qualities and productivity. Finally, studying the Leonurus japonicus' s qualities based on the optimized technology to establish the corresponding draft of Four System Leonurus japonicus' s quality standard, reveal the Four System Leonurus japonicus' s meaning deeply.By measuring some of the main indexes, the quality differences between different areas, different harvesting time and different drug sites were determined, which provided scientific basis for the cultivation and clinical application of Motherwort.Methods1.Harvesting period, origin and drug site' s studyThe content of alkaloids and flavonoids in 17 different habitats, different harvesting stages and different medicinal herbs were determined by HPLC.The contents of the same components in different areas were different. In the determination of different parts,The contents of alkaloids and alkaloids were higher than that of stem, and the chromatograms of 17 producing areas were analyzed systematically. The method of determination of alkaloids and flavonoids was established by methodological study.2.Processing technology' s studyFour kinds of auxiliary materials were used as the evaluation index. After the four kinds of auxiliary materials were sprinkled and then steamed, the samples were tested and the uniform design test was carried out.The solution of the four kinds of auxiliary materials was carried out.Of the simplified, effective alkaloids and flavonoids as far as possible to retain the problem,a better processing technology.3.Study on the fingerprintThe fingerprints of Tetradis mandshurica were established by HPLC with the method of using eugenolus officinalis as the reference peak, and the common peak information was obtained. The similarity analysis of common peak information was carried out by using the similarity degree evaluation system of traditional Chinese medicine fingerprint similarity evaluation system. And its products four system of Leonurus japonicus.4.The quality standard' s studyThe contents of traits, microscopic, thin layer, water, ash, alkaloid and flavonoids were determined according to the detection method of Chinese Pharmacopoeia 2015 edition. The evaluation of different batches of motherwort was carried out. Finally, according to the experimental results, the development of the corresponding vinegar four system of motherwort quality standards. Finally, based on the experimental data to establish the Four System Leonurus japonicus' s corresponding quality standard draft.Results1.Harvesting period, origin and drug site' s study In general, the contents of alkaloids and flavonoids in 17 batches of medicinal herbs were significantly different. The reason for the difference was related to the harvesting method, harvest season and time, storage and other factors.Analysis of the two content combined with medicinal resources to consider, the best medicinal parts for the leaves, the best harvest period in mid-June.2.Processing technology' s studyThrough the investigation of key steps in the processing of Tetradis carbohydrate, the infiltration time, steaming time and salt, alcohol, ginger and vinegar were optimized. The results showed that the optimum process was as follows:steaming for 0.5h,infiltration time of 1h,the amount of excipients was 3g of ginger juice per 100g,10g of wine,10g of salt and 2g of salt. And the pilot experiment was carried out.The results show that the process is reasonable and feasible.3.Study on the fingerprintThe fingerprints of Leonurus japonicus and its products were established. There were 23 peaks of HPLC fingerprints of 17 herbs and four motherwort, and the fingerprints of motherwort slices were analyzed and compared. The similarity of map shows that there are some differences in the different harvesting period, and the difference is obvious, but the composition is basically the same, but the content of some components is different.4.The quality standard' s studyTest the Four System Leonurus japonicus;s shape, microscopy, the Thin Layer Chromatography(TLC),the moisture content, the total ash and the amount of its alkaloids to analyze different batches' Four System Leonurus japonicus;s qualities and determine the Four System Leonurus japonicus' s quality standar draft.Conclusion1.Harvesting period, origin and drug site' s studyIn this study, the contents of alkaloids and flavonoids in Herbamongolicum officinalis were studied systematically. The results were as follows:the identification of medicinal materials, the evaluation and rational utilization of resources, the clinical use of medicinal materials and the later research. The scientific basis.2.Processing technology,s studyOptimize the vinegar four system of motherwort processing technology, standardize the process of processing indicators,to enhance product quality and production efficiency at the same time, for large-scale production provides a theoretical basis and basis.3.Study on the fingerprintThe HPLC method has the advantages of high sensitivity and strong specificity.HPLC fingerprint and alkaloid content can fully reflect its intrinsic quality and provide scientific basis for the quality control of Coptis chinensis.4.The quality standard' s studyDraft the Four System Leonurus japonicus' s quality standard including the demand of identification in appearances,TLC' s requirement, the moisture content lesses than or equals 12.0%,the total ash lesses than or equals 11.0%,the content of Stachydrine hydrochloride lesses than or equals 0.50%,the content of Leonurus hydrochloride lesses than or equals 0.05%,the content of quercetin and kaempferol lesses than or equals 0.10%.
