Objective: In recent years,with the improvement of living standards,increasing bad habits,the cervical spodylosis is increasing year by year,and the cervical spondylotic radiculopathy(CSR)incidence ratio is particularly prominent,seriously affecting people's health;therefore,the cervical spondylotic radiculopathy's prevention is the focus of current medical research where.The study confirmed that the JAK-STAT/SOCS signaling pathway mediated the pain mechanism of cervical spondylotic myelopathy.The purpose of this study was to investigate the effect of electroacupuncture at Siguan Points on the interaction of JAK-STAT/SOCS signal in the model of cervical spondylotic radiculopathy type rats,,acupoints from distal to forward angle,to reveal the mechanism of acupuncture treatment of cervical spondylotic radiculopathy.Methods: SPF 60 Wistar rats,male and female of each half,weight250±20g,let the rats for 1 weeks to adapt to the environment,using a completely randomized block design,60 rats were divided into 3 groups:blank control group(blank group),CSR model group(model group)and electroacupuncture group(EA group,n = 20).Establishment of animal model of cervical spondylosis in rats.Operation 3 days after modeling through the behavior observation confirmed that the model successfullyreproduced,and fixed respectively to be fixed after Electro Acupuncture Acupuncture Siguan Points and electroacupuncture treatment,the control group and the model group only in the same conditions.After 7 days the rats were sacrificed,the pressure at(segment C6-T1)of spinal cord and nerve root tissue by immunohistochemistry and reverse transcription polymerase chain reaction(RT-PCR),CO immunoprecipitation(CO-IP)combined with Western blotting(Western Blot),to detect the expression of JAK1,STAT3,SOCS1 mRNA,SOCS3mRNA SOCS1-JAK1,SOCS3-STAT3 protein,observe the effect of Electroacupuncture of Siguan Points of JAK1,STAT3,SOCS1 mRNA,SOCS3mRNA,SOCS1-JAK1,SOCS3-STAT3 content and expression of the content.Results:(1)Immunohistochemistry: compared with the blank group,the expression of JAK1 in the model group,STAT3 significantly increased,the difference was statistically significant(P<0.05);compared with the model group,EA group,STAT3 JAK1 expression was significantly reduced,the difference was statistically significant(P<0.05).(2)RT-PCR detection: compared with the blank group,the expression of SOCS1 mRNA in the model group,SOCS3 mRNA significantly increased,the difference was statistically significant(P<0.05);compared with the model group,EA group,SOCS3 mRNA SOCS1mRNA expression increased significantly,the difference was statistically significant(P<0.05).(3)CO-IP detected in the 3 groups,there was expression of JAK1SOCS1 in washing liquid immunoprecipitation;the expression of SOCS3STAT3 in washing liquid immunoprecipitation.It is indicated that the binding protein of SOCS1-JAK1 and SOCS3-STAT3 in the tissue undertest,the reaction between JAK1 and SOCS1,STAT3 and SOCS3 may be directly affected.Conclusion: 1.JAK-STAT signaling pathway and its inhibitor SOCS may interact with each other through direct binding.2.Electro acupuncture Siguan Points can regulate the expression of JAK,STAT,SOCS,thereby regulating the JAK-STAT/SOCS signaling pathway.3.Electro acupuncture Siguan Points may be to improve the expression of SOCS protein,inhibit the JAK-STAT signal transduction pathway to achieve the purpose of treatment of CSR root pain.
