Abstract/Summary:
Object:1.Observation of herb-medicine-cake-separated moxibustion on atherosclerosis(AS)rabbit liver tissue of nuclear receptor LXRα m RNA gene expression changes;2.To observe the expression changes of LXRα protein in the liver tissue of AS rabbits;3.Verify herb-medicine-cake-separated moxibustion by AS rabbit regulate LXRα in reverse cholesterol transport,and promote the role of anti-AS.Methods:1.Feeding Preparation of AS rabbit model of success through high cholesterol,with its herb-medicine-cake-separated moxibustion for the intervention,simvastatin as a positive control,continuous treatment for 4 weeks;2.The morphological structure of rabbit aortic wall and liver was observed under optical microscope;3.Liquidscintillation counting was used to detect the changes of serum lipid and the content of cholesterol in rabbit liver;4.Real-time fluorescence quantitative polymerase chain reaction(RT-q PCR)was used to detect the m RNA of nuclear receptor LXRα in liver tissue;5.The nuclear receptor LXRα protein in rabbit liver was detected by Western blotting.Result:1.Level rabbit lipids: compared with the normal group,the model group erum TC and low-density lipoprotein levels were higher,but the serum high-density lipoprotein was lower(P <0.01);Compared with the model group,the treatment group serum TC and low-density lipoprotein levels were significantly reduced,but the serum high-density lipoprotein increased(P <0.01);2.The levels change of cholesterol,low density lipoprotein and high density lipoprotein in rabbit liver tissue of each group: compared with the normal group,the model group contents of cholesterol and low density lipoprotein were higher,but the content of high density lipoprotein reduced;Compared with the model group,the changes in the treatment groups were just the opposite 3.The expression of LXRα protein in the rabbit liver of each group: compared with the normal group,the expression of LXRα protein in the model group reduced accordingly;Compared with the model group,the expression of LXRα protein in the moxibustion group and the drug group increased accordingly,and the increase of the moxibustion group was more obvious.4.The expression of LXRα protein and m RNA in the rabbit liver of each group: compared with the normal group,the LXRα protein and m RNA expression in the liver of the model group reduced(P<0.01 or P <0.05);Compared with the model group,the LXRα protein and m RNA expression in the liver of the treatment groups showed an increasing trend,the difference was statistically significant(P<0.01 or P <0.05).Conclusion:1.The AS rabbits can be successfully produced by using the high fat diet method in this study.2.The herb-medicine-cake-separated moxibustion method had a decreasing effect on TC and LDL in serum and liver tissue of AS rabbits,and had a rise effect on HDL.3.The herb-medicine-cake-separated moxibustion can activate LXRα m RNA and protein expression,promote cholesterol reverse transport,thereby inhibiting or slowing the formation of atherosclerosis.
