Abstract/Summary:
Object:1.Observation of herb-medicine-cake-separated moxibustion on atherosclerosis(AS) rabbit liver cells, fat cells, peritoneal macrophages and peripheral blood mononuclear cells of ATP-binding cassette transporter A1(ABCA1)expression changes; 2.Determination of herb-medicine-cake-separated moxibustion on AS rabbit liver cells, fat cells and peritoneal macrophages [3H] cholesterol transferred out of the rate of change;3.Verify herb-medicine-cake-separated moxibustion by AS rabbit regulate ABCA1 in reverse cholesterol transport, and promote the role of anti-AS.Methods: 1.Feeding Preparation of AS rabbit model of success through high cholesterol, with its herb-medicine-cake-separated moxibustion for the intervention,simvastatin as a positive control, continuous treatment for 4 weeks;2. Cholesterol was measured using enzyme immunoassay and lipid changes rabbits and rabbit liver, aorta,adipose tissue;3. Under an optical microscope rabbit aortic wall morphology;4.Liquid scintillation counting was used to detect rabbit liver cells, fat cells and peritoneal macrophages [3H] cholesterol feeding rate;5. Expression by flow cytometry rabbit liver cells, fat cells, peritoneal macrophages and peripheral blood mononuclear cells of ABCA1.Result:1. Level rabbit lipids: compared with the model group, the treatment group serum TC, TG, LDL-C, Apo-B levels were significantly reduced(P <0.01), HDL-C,Apo AI levels were increased(P <0.05 or P <0.01).2. Cholesterol rabbit of the organizations: Compared with the model group, the treatment group aorta, adipose tissue, liver tissue was significantly reduced cholesterol content(P <0.01).3. Rabbit each cell [3H] cholesterol feeding rate: Compared with the model group, the treatment group peripheral mononuclear cells and peritoneal macrophages, adipocytes cholesterol feeding was significantly increased(P <0.01), and hepatic cholesterol turn out rate decreased(P <0.05).4. Expression rabbit each cell of ABCA1: compared with the model group, the treatment group peripheral mononuclear cells and peritoneal macrophages, expression of ABCA1 adipocytes were increased(P <0.05 or P <0.01),and hepatocyte ABCA1 protein expression decreased significantly(P <0.01).Conclusion:1.Herb-medicine-cake-separated moxibustion on AS rabbit lipid metabolism has a good regulation.2. Herb-medicine-cake-separated moxibustion regulated expression of ABCA1 by peripheral cells, and elevated cholesterol in peripheral cells turn out rate, promote RCT to exert its anti-AS effect.
