Objective1 To establish the optimal operating conditions of compound decoction extracted by two fried normal pressure decocting machine.2 To set up a quality control system of compound decoction extracted by two fried normal pressure decocting machine under the optimal operating conditions.MethodsThe content of the main active ingredients as the inspection target,utilizes the orthogonal experimental method to obtain the optimum conditions for compound decoction;TLC and HPLC were used to control the quality of the main active ingredients for compound decoction;Compound decoction was prepared in the optimal conditions,and the content change of the main active ingredients was observed within 7 days at room temperature and avoided light;meanwhile,microbial limit test was carried out.Results1 This experiment takes the content of matrine as the inspection target of Qingshiwaixi Decoction,takes the content of protocatechualdehyde as the inspection target of Huoxuezhitong Decoction,through the orthogonal experimental method to analyze the effect of the four factors,including the decoction time in the first time,the decoction time in the second time,the immersion time,the volume of water,and to obtain the optimum conditions for compound decoction.The optimum conditions:the decoction time in the first time is 30min,the decoction time in the second time is 20min,the immersion time is 30min,and the volume of water is 1200ml.2 This experiment identify the species of traditional Chinese medicine in the decoction by TLC,including Fructus Cnidii,Cortex Phellodendri,Fructus Gardeniae in Qingshiwaixi Decoction,and Radix Salviae Miltiorrhizae,Radix Angelicae Sinensis,Radix Notoginseng,Rhizoma Corydalis,Radix Aucklandiae in Huoxuezhitong Decoction.The spots in TLC were clear,negative control without interference.The selected method is good in separation,strong in specificity,and provides a reliable experimental data for the quality control of the selected decoction.3 This experiment determines the content of the main active ingredients in the decoction by HPLC,including matrine,gardenoside in Qingshiwaixi Decoction,and protocatechualdehyde,ferulic acid in Huoxuezhitong Decoction.The selected method has high sensitivity,strong specificity and good reproducibility,and provides a scientific experimental data for the quality control of the selected decoction.4 The content of matrine and gardenoside in Qingshiwaixi Decoction showed 1.85%?1.10%decrease in the 7th day.The content of protocatechualdehyde and ferulic acid in Huoxuezhitong Decoction showed 2.03%.1.88%decrease in the 7th day;the results of microbial limit test met the sanitary standards of drugs.ConclusionThe results show that this method is simple,highly sensitive,highly repeatable and specific,and can be used for quality control of compound decoction extracted by two fried normal pressure decocting machine.Compound decoction could be kept stably at room temperature and avoided light lasting 7 days after it was extracted by two fried normal pressure decocting machine.
