| The anxiolytic effect of huanglianejiao (HLEJ) decoction in mice preliminarily investigated by means of behavioral methods for the first time. The side-effect was also investigated in the work. The results shows that the HLEJ decoction at the doses 5.0g kg-1 prolonged the duration of immobility of the mice in the tail suspension test ,the spontaneous motor activity and reduce the ratio attact behavior in the stimulated by electric shock test significantly. It changed the roto-rod performance but the result wasn't significantly. All these findings indicated that HLEJ decoction exhibits anxiolytic-like effects and doesn't influence the muscle and motor coordination.On the base of this experiment, with the aid of orthogonal design, we decided the better prescription is that huanglian12g, huangqin6g, baishao6g, ejiao9g.By using berberine hydrochloride, baicalin as quality control criteria traditional extraction process of HLEJ decoction was optimized with the aid of orthogonal design. Quantitative assays were performed on active components of berberine hydrochloride, baicalin by HPLC. They had good linearity within the ranges of 0.84~3.36ug (r=0.9993) ,0.372~ 1.860ug (r=0.9992) respectively. The suitable conditions as follows: adding lOtimes amount of water, decocting for 2times,60min each time. |
