Purpose:Gastric cancer(GC)is a common malignancy of the digestive system worldwide.Patients with GC often show a poor prognosis because it is hard to diagnose GC at a early stage.Therefore,it is crucial to develop novel diagnosis markers for this large group of patients.Previous studies reported that the abnormal expression of KIF18 A involves in the tumorigenesis and development of loads of cancers.However,the relationship between KIF18 A and GC remains unclear.In this study,we aimed at analyzing the expression level of KIF18 in GC,and the relationship between KIF18 and the prognosis of GC patients.Methods:Real-time PCR analysis was used to detect KIF18 A m RNA levels in 20 pairs of gastric cancer and corresponding normal gastric tissues.The expression of KIF18 A protein was detected using Immunohistochemistry in 129 gastric cancer cases and 23 normal mucosa to evaluate the KIF18 A prognostic significance,Kaplan-Meier method and Cox's proportional hazards model were used in survival analysis..Results:The expression of KIF18 A m RNA in 16 gastric tissues was obviously lower than that in corresponding normal tissues among total 20 pair tissues.The result of Immunohistochemistry shows that KIF18 A positive rate in gastric carcinoma was significantly lower than that in normal control tissue(P=0.041).In addition,the expression of KIF18 A may related to TNM staging of gastric cancer(P=0.032)and depth of invasion(P=0.008).Kaplan-Meier analysis revealed that patients with low expression of KIF18 A associated with shorter overall survival(P=0.002).Furthermore, the data of univariate Cox regression model indicated that the expression of KIF18 A,TNM stage,differentiation,gastric wall invasion,lymph node metastasis related to overall survival.Finally,multivariate Cox analysis suggested that the expression of KIF18 A,differentiation and TNM stage could be an independent prognostic factor of gastric cancer to predict overall survival of patients.Conclusions: The expression of KIF18 A was significantly lower in gastric cancer than that in normal gastric tissues,and the overall survival markedly shorten in patients with low expression of KIF18 A than those with high expression of KIF18 A,the lower expression of patients' prognosis is poor.KIF18 A may be a crucial factor of tumorigenesis in gastric cancer and may be a potential independent prognostic factor for gastric cancer.With the deepening of the research on KIF18 A gene,it may be as a new biological marker has an important significance for early diagnosis of gastric cancer and prognosis prediction.
