The Expression Of Tmprss 4 In Gastric Cancer Tissues And Its Clinical Significance

Background and purpose Gastric cancer is a highly malignant disease,and is considered by genetic,epigenetic,and environmental impact caused by many factors.The low diagnostic and resection rate is the main cause of poor prognosis in patients with gastric cancer.Many biological markers used to judge the diagnosis?treatment and prognosis such as serum carcinoembryonic antigen(CEA)and cancer of the stomach tissue HER2 expression has been suggested as a gastric cancer markers and used in clinical practice.However,their low sensitivity and specificity limits its application.Diagnostic markers,prognostic factors and the lack of effective therapeutic targets is still contribution to the poor efficacy of clinical diagnosis?treatment and prognosis of gastric cancer.Therefore,further research on the pathogenesis of GC to discover new molecular markers for diagnosis and prognosis of gastric cancer for the improvement of gastric cancer patients quality of life and survival is a problem which needs to be settled.Explore Transmembrane serine protease 4(Transmembrane protease,serine 4 TMPRSS 4)expression level in gastric malignant tumor and its relationship with clinical pathology characteristic GC patients,analysis TMPRSS 4 protein expression levels and the prognosis of patients with GC survival,in order to discover new biological markers of GC.Methods: RT-qPCR methods were used to detect the expression of TMPRSS 4 mRNA in 41 cases of the Fresh tissue samples(Cancer tissue and normal tissue adjacent to carcinoma)of GC patients who confirmed of GC(Gastroscope pathological biopsy or postoperative pathology)and accept surgical treatment in Anhui provincial hospital affiliated to Anhui medical univarsity;immunohistochemical methods were used to detect the expression of TMPRSS4 in 115 cases of GC.Analysis of the relationship between its expression differences and pathology in GC patients,as well as TMPRSS 4 protein expression levels in gastric malignant tumor on the survival prognosis of patients with GC.Results 1.The results of RT-qPCR showed that the expression of TMPRSS 4 mRNA in gastric cancer tissues was significantly higher than that in the adjacent tissues,and the difference was statistically significant(0.86+0.31vs0.41+0.15,P<0.05).2.Immunohistochemistry results showed that the expression of TMPRSS 4 in gastric cancer tissues was higher than that in benign tissues(70.4% vs 19.5%,P<0.001.)3.Clinical data analysis: the over expression of TMPRSS4 was significantly correlated with the degree of differentiation,lymph node metastasis and high TMN staging of gastric cancer(P= 0.020,P < 0.001,P=0.066).TMPRSS4 is an independent prognostic factor for overall survival and disease-free survival in patients with gastric cancer.Conclusions The high expression of TMPRSS 4 and are closely related to clinical pathological features in patients with gastric cancer and their prognosis.TMPRSS4 may be used as a conventional gastric cancer biomarkers can predict tumor invasion and metastasis in a certain extent.
