ObjectiveTo observe the effect of Qingchang Jiedu Decoction, on the expression of IL-6 and NF-?Bp65 of rats in UC damp heat syndrome model. To examine the effectiveness of treating UC through the methods of clearing heat and promoting diuresis,promoting blood circulation to remove meridian obstruction. To provide the fundamental basis of clinical use of Qingchang Jiedu Decoction.MethodsThe successful model of 50 SPF SD rats(half male and half female), were randomly divided into model group, western medicine, and high-dosage, medium-dosage and low-dosage of Qingchang Jiedu Decoction groups (together called the Chinese medicine arm).n= 10; Another 10 normal rats as a control group.We built the rats UC damp heat syndrome model using compound inducements(damp-warm environment, high-fat and high-carbonhydrate diet, and clyster with 50% TNBS ethanol solution). Once samples got UC, in Chinese medicine arm, rats were given intragastric administration with respective dosages of Qingchang Jiedu Decoction. In Western medicine group, rats were given intragastric administration with SASP. And rats in control group and model group were given intragastric administration with physiological saline of the same volume as Chinese medicine arm. Intragastric administrations were given once a day for two weeks. After that, rats were killed. Then the expression of IL-6 and NF-?Bp65 protein DNA in rats'colon tissues were evaluated through the methods Immunohistochemical staining SP and The average optical density. And the pathomorphological change of rats'mucous membrane of colon tissues were examined through slicing.Results1.General status:Rats got better appetite, lustrous coat and formed feces. Rats did not have loose, mucous or bloody purulent stool. Rats' weight increased. The increasing was significant in Chinese Medicine (CM) high-dosage group and Western Medicine (WM) group (P<0.05 after 7 days of treatment, and P<0.01 after 14 days of treatment).2.The pathomorphological change of rats mucous membrane of colon:Qingchang Jiedu Decoction decreased the damage of mucous membrane of colon tissues and the pathological histology scores. The scores of CM low-dosage group were comparable to that of model group. The scores of CM medium-dosage group were better than that of model group. Rats in CM high-dosage group and WM group had some inflammation in lamina propria of colon mucous membranes, but no thickening in muscularis mucosae or congestive edema in submucosa. The scores of CM high-dosage group were close to that of WM group.3.NF-?Bp65 in rats mucous membrane of colon:Compared to model group, the decreasing of contents of NF-?Bp65 in rats colon tissues in CM high-dosage group and WM group was highly significant (P<0.01); the decreasing in CM medium-dosage group was significant (P<0.05); and the decreasing in CM low-dosage group was not significant.4.IL-6 in rats mucous membrane of colon:Compared to control group, the increasing of contents of IL-6 protein in rats colon tissues in model group was highly significant(P<0.01). Compared to model group, the decreasing of contents of IL-6 protein in rats colon tissues in CM high-dosage and WM group was highly significant (P<0.01); the decreasing in CM medium-dosage group was significant (P<0.05); and the decreasing in CM low-dosage group was not significant.Conclusions1.The increasing of NF-?Bp65 and IL-6 in mucous membrane of colon might be one of the pathogenesis of ulcerative colitis.2.A possible mechanism of treating ulcerative colitis damp heat syndrome by Qingchang Jiedu Decoctionwas that Qingchang Jiedu Decoction performed against inflammation and protected mucous membrane of colon through decreasing the levels of NF-?Bp65 and IL-6.3.By observing the general status of rats after treatment, the pathomorphological change of rats mucous membrane of colon and the levels of NF-?Bp65 and IL-6, we confirmed the effectiveness of Qingchang Jiedu Decoction.
