[Objective]Through the experimental to research the influence of Qing-chang Jiedu Decoction to the index IL-8 and TNF-?, the inflammation of Colonic mucosa, in model rats of The Ulcer Colitis Rdhs. To discuss the mechanism of the Decoction treat The Ulcer Colitis, and provide new ideas for the treatment in traditional Chinese medicine of The Ulcer Colitis.[Method]The SD male rats of SPF class use the compound method which are TNBS (three nitrobenzene sulfonic acid)/ethanol+diet+circumstance to induce the model rats of The Ulcer Colitis Rdhs. To observe the pathological changes of colonic mucosa in experimental rats through the HE staining method, to check the expression of IL-8 in colonic mucosa of experimental rats through the immunohistochemical method and the content of TNF-? in serum of experimental rats through the ELASA method. After the successful experimental of rats, the number of female and male rats was equal, divided the rats into six group which are blank group, model group,positive control group, high, medium and low dose group of Traditional Chinese medicine.[Result](1) Histopathology of colonic mucosa in experimental rats:?Compared with the blank group:The pathological score of the model group was significantly higher than that of the blank group, and the difference was statistically significant(p<0.01). ?Compared with the model group:The pathological score of the traditional Chinese medicine high dose group was significantly lower than that of the model group, the difference was statistically significant(p <0.01);The chinese medicine low and middle dose group have pathological score lower, but the difference is not statistically significant (p>0.05);The pathological score of rats in western medicine group was significantly lower, the difference was statistically significant(p<0.01). ?Compared with the western medicine group:The chinese medicine high dose group and the middle dose group rats pathological score change is not obvious, the difference was not statistically significant;The pathological score of the low dose group of Chinese medicine group was significantly higher, the difference was statistically significant(p<0.05).(2) Expression of IL-8 in colonic mucosa of rats:?Compared with the blank group:The average light density of IL-8 in the model group was significantly higher, and the difference was statistically significant(p< 0.01). ? Compared with the model group:The average light density of IL-8 in the colon mucosa of the traditional Chinese medicine group was significantly lower than that of the model group, the difference was statistically significant(p< 0.01);?Compared with the western medicine group:The average light density of IL-8 in the high dose group of traditional Chinese medicine group had no significant change, and the difference was not statistically significant;The changes of average light density of IL-8 in the colon mucosa of Chinese traditional medicine middle and low dose group, the difference was statistically significant (p<0.05).(3)The content of serum TNF-?:?Compared with the blank group:The content of TNF-? in serum of model group was significantly higher than that of the blank group, and the difference was statistically significant (p<0.01). ? Compared with model group:The content of TNF-? in serum of the rats of the traditional Chinese medicine groups of was significantly lower than that in the model group, the difference was statistically significant (p<0.01);The content of TNF-? in serum of rats in western medicine group was significantly decreased, the difference was statistically significant(p<0.01). ?Compared with the western medicine group:There was no significant difference in the serum TNF-?levels of the traditional Chinese medicine group and the middle dose group, the difference was not statistically significant;The content of TNF-in the serum of rats in the low dose group of Chinese medicine was significantly different, and the difference was statistically significant(p <0.01).[Conclusion](1)Qingchang Jiedu Decoction can reduce the expression of IL-8 in colonic mucosa and the content of TNF-a in serum, and there was no significant difference between the high dose group and Western medicine group.(2) Qingchang Jiedu Decoction can alleviate the pathological damage in rats,and the therapeutic effect was positively correlated with the dose of traditional Chinese medicine.
