This paper was mainly research on changes of chemical composition, hot property, efficacy and toxicity of acontum fischeri during processed into Heishun Pian. Aimed at enriching research of "the attenuated deposit efficiency" principle of acontum fischeri during the preparation process, ensureing the safety and efficiency of Fuzi.It is hoped that the study results can help heritage and protect the traditional processing skill of acontum fischeri, offer certain basic guidance for new processing technology or preparation methods of acontum fischeri, provide new ideas for research of herbal preparation process at the same time.Chemical composition determination was conducted by HPLC to check contents of four kinds of water-soluble component:dopamine hydrochloride, uracil, uridine, guanosine and six kinds of ester alkaloids:aconitine, mesaconitine, hypaconitine, benzoylaconine, benzoylmesaconitine, benzoylhypaconine from lyophilized powder and aqueous extract solution of Heishun Pian after various processing methods (Nifuzi, Danfuzi, Zhufuzi, Bingfuzi, Piaofupian, Zhengfupian, Heishunpian) and content of bittern in the water extract sample was determined by the titration method. The results showed that the contents of uracil, uridine, guanosine were the highest in Nifuzi and the highest dopamine hydrochloride content in the Bingfuzi; the content of four kinds of water-soluble component after the rinsing was reduced significantly; but in aqueous extracts of Heishun Pian after various processing methods except ester alkaloids, compared with the original medicinal materials the bittern and four water-soluble components' contents were decreased in the water extract and the change rules of bittern and water-soluble component contents were similar.Hot property experiments were performed by intragastric administration with aqueous extracts of Heishun Pian after various processing methods in rats. Observe and record changes of vital signs of rats in the experimental process and determine biochemical indicators related to hot property of acontum fischeri, and according to differences between each biochemical indicator to analyze variation law of hot property during process of acontum fischeri being processed into Heishun Pian. Study results showed that the hot property of acontum fischeri in nature would be affected by addition of bittern, reduction and transformation of ingredients and the preparation process reduced acontum fischeri's hot property.Specific to actions of supplementing fire and strengthen yang and expelling cold to relieve pain, constipation due to yang-deficiency experiment, low-temperature swimming experiment and anti-inflammation and analgesia experiments were conducted to explore changes of efficacy of Heishun Pian in the process of preparation. Study results found that the addition of bittern would influence the action of warming spleen-yang and reduce analgesic action for inflammatory pain in the preparation process; the preparation process can make contribute to the action of supplementing fire and strengthen yang, but the action of expelling cold to relieve pain of acontum fischeri after processing would be weaken.Acute toxicity and heart toxicity experiments were carried out through aqueous extracts and alcohol extracts of Heishun Pian samples after various processing methods, results showed that aqueous extract samples without or with little bittern had no toxicity, but aqueous extract samples with bittern contain obvious toxicity and the more toxicity with the increasing content of bittern. Toxicity in various samples of alcohol extracts was the synthesis of bittern and the aconitine compositions, the higher content of bittern, the bigger toxic of alcohol extracts with different samples; the toxicity of acontum fischeri after processing would be weaken.The above results show that the preparation process of Heishun Pian is not only the process of detoxification, but also would produce certain influence on actions and hot property; which told that the study of Chinese herbal process should not be limited by the researches pattern of "the beginnings and endings". The research provide more research data and the foundation acontum fischeri's processing methods through the above a series of studies.
