Siwu decoction is honored as effective medicine for replenishing blood and regulating menstruation. Siwu mixture is the improved achievement fromtraditional Siwu decoction. Quality controlling method of unique importance of class origin is unilateral wh ich cannot reflect the function and preparationprocess of pharmaceutical preparation. This th esis combines the fingerprint and efficacy of midbody from Siwu mixture's extraction and purification pro-cess to establish profile-effect relationship, to control the preparation proces seffectively.Objection:This thesis represents chemical information and pharmacodynamic effects of midbodies from Siwu mixture's extraction process, and establishes the profile-effect correlation, which provides reliable and objective foundation for controlling and evaluating standards of Siwu mixture's preparation process.Method and Results:In the first part, the author makes use of HPLC to establish the fingerprint of midbodies ?distilled water, water decoction, ethanol supernatant, alcohol sediment? which are from Siwu mixture extraction and purification process, and uses fingerprints characteristic peaks of different extraction products as variables to do system collection analyses. It is showed that when euclidean distance is 5, the midbodies from Siwu mixture extraction and purification process can be differentiated clearly.In the second part, the author measures midbodies'relevant pharmacodynamical parameter. It shows that when mice suffer from acute blood stasis caused by adrenal gland ice-water bath, compared with model set, except Siwu alcohol sediment group, all the medicated groups can lower plasma viscosity and blood viscosity under different shear rate, and Siwu water decoction group and ethanol supernatant group function better. When APH and CTX lead to blood deficiency in mice, compared with model set, all the medicated groups can rise index of WBC, HGB, RBC, HCT to some extent, and it can also rise index of liver and thymus, and Siwu ethanol supernatant group has best efficacy. When oxytocin leads to dysmenorrhea of mice, compared with model set, all the medicated groups can rise PGE2 content and lower PGF2? in blood viscosity, Siwu water decoction and alcohol sediment groups have much better efficacy.In the third part, the author does PCA ?principal component analysis? in pharmacodynamics index through blood stasis, blood deficiency and dysmenorrhea model experiments, and simplifies 30 fingerprint peaks. Through gray relational grade method, the author also analyzes footprint characteristic peaks and pharmacodynamics integrated parameter. Finally the author finds in 4 extractions footprint characteristic peak 2,5,7.8,11,12,13,21 contributes most. By comparison, it shows that peak 2 is catalpol, peak 7 is gallate, peak 21 is paeoniflorin.Conclusion:This thesis does spectrum-effect study on distilled water, water decoction, ethanol supernatant, alcohol sediment from Siwu mixture's distillation, water-extraction and alcohol precipitation process. Based on all the studies, the conclusion is what does well in promoting blood circulation is the high polarity constituents from decocting technology. And what does well in enriching blood and regulating menstruation is the low polarity constituents from alcohol precipitation process. The author also finds that what contributes most to fingerprint peak is catalpol, gallate, paeoniflorin and some other unknown compounds, but not ferulic acid. It provides new thoughts for scientific evaluating standard between quality and efficacy of compound preparation process.
