Chinese medicine theory holds that Peucedanum praeruptorum classified as slightly cold, bitter, acrid. In the lungs,spleen,liver.With the evacuation of wind-heat,qi-descending and reso lving phlegm.In this paper imperatorin resource distribution, chemical com position,pharmacological effects,quality standards review, based on the effective fraction of Peucedanum praeruptorum extraction process was optimized. the total extraction rate of coumarin and the dry extraction as indicators,ethanol concentration,ethanol volume, extraction time, extraction times for orthogonal test fac tors were studied to determine the optimum extraction condition s:8 times 75%ethanol, extracting 3 times 0.5 hours. And the be st technology was verified.Total coumarin content of the eluate as a indicator.The tech nology which enrich and purify effective fraction of Peucedanum praeruptorum for macroporous resin were investigated,the extrac tion liquid was concentrated to 0.1 g per 1 ml amount of raw me dicinal herbs,0.2 g per 1 ml amount of raw medicinal herbs and 0.5 g per 1 ml amount of raw medicinal herbs, then put it aside and found a large number of insoluble matter. By ultraviolet sp ectrophotometry, with a total loss of coumarin content 21.0%,4 7.97%,63.01%.The extraction liquid was concentrated no alcohol,then to 0.5 g per 1 ml amount of raw medicinal herbs. Content of total cou marin and recovery rate of dry extraction as indicators. It rese arch on reagent type,reagent dosage,solvent volume three factor s by orthogonal test method to determine the optimum extraction conditions:0.5 time the amount of n-butanol 3 times.The best te chnology was verified.It obtain the effective fraction of Peuce danum praeruptorum.Using 7-hydroxycoumarin as reference substance by UV spec trophotometry,it determine the total coumarins content of effec tive fraction for Peucedanum praeruptorum. The detection wavelen th was 321nm.The methodological study were investigated,the lin ear equation was Y=0.0928X+0.077(R2= 0.9992).The concentration on of effective fractions between 1.952-7.808μg/mL show a goo d linear relationship with absorbance. Recovery rate was 99.745%, RSD=0.55%(n=6).To develop a high performance liquid chromatography method for determinating the contents of praeruptorin A,praeruptorin B in the effective fractive.Standard curve for Praeruptorin A was Y=464.42X+0.5438 (R2=0.9998,n=5). The linear response ranged from 0.32-6.4μg for praeruptorin A between weightand peak are a. Determination the content of praeruptorin A in the effective fractive content was 77.38mg per g. The rate of transfer was 83.55%for praeruptorin A in extraction. Average recovery was 98.92%, with RSD=1.6%(n=5). Standard curve for Praeruptorin B was Y= 364.86X-2.2932, R2= 0.9998 (n=5). The linear response ran ged from 0.4-3.6μg for praeruptorin B between weight and pea k area. Determination the content of praeruptorin B in the effec tive fractive content was 23.90mg per 1g. The rate of transfer was 83.24%for praeruptorin B in the extraction. Average recover was 99.66%,with RSD=1.5%(n=5).Systematically studying on the components of coumarin using the fingerprint by HPLC-UV method, to establish the fingerprint total model for Peucedanum praeruptorum. It evaluate the quality of the Peucedanum praeruptorum around the world application " Chinese chromatographic fingerprint evaluation system". Similari ty results show that:extractions of various medicinal herbs and reference Peucedanum praeruptorum similarity were between 0.6-0.93,initially identified A total of 4 fingerprint peaks.
