Objective:According to the theory of the five elements and the theory of Tibetan images,from the spleen,lung gold and liver wood of the five elements of the principle of grams and the overall view of the viscera,observed with spleen Qi,spleen and Yangfei,Shugan spleen and lungs three different Treatment of Pulmonary and Spleen Deficiency Syndrome Rat Models by Treatment of Traditional Chinese Medicine(The prescriptions were flavored JWYG San?HWYG San and CWYG San).Explore the efficacy and the mechanism of different treatment on immune function and neurotransmitters in rats with deficiency of spleen and spleen which treatment provides new ideas and basis.Methods:46 young male Wistar rats of SPF grade,3-4 weeks of age,weighing about 105 g were randomly divided into two groups:normal group(n=10)and Zaomo group(n=36).(1)Three days to adapt to rearing,(in addition to the normal group)the Zaomo group smoked,papain fog and reserpine 0.2mg/kg intramuscular injection,modeling 30 days.On the 31 st day,2 rats were randomly selected from the normalgroup and the Zaomo group.Pathological changes were observed in lung,stomach and small intestine.(2)After successful operation,the Zaomo group was divided into four groups: model group,Jianpi Yiqi group(abbreviated as Jiawei group),Jianpi Yangfei group(referred to as Huangwei group)and Shugan Jianpi Yangfei group(abbreviated as Chaiwei group),8 rats in each group.The rats in the normal group and the model group were given normal saline once a day,and the Jiawei group was given JWYG San,the Huangwei group was given HWYG San,the Chaiwei group was given CWYG San.The group was divided into three groups according to the concentration of1ml/100 g,which lasted for 18 days.The body weight,fur,food intake,stool condition,mental state,activity and so on were observed.At the end of the 7th week,the rats were sacrificed at the end of the 7th week,and the supernatant was taken from the abdominal aorta.The supernatant was taken from the blood of the rat brain.The supernatant was centrifuged and the supernatant was collected by Enzyme Linked Immunosorbent Assay(ELISA).The levels of neurotransmitter norepinephrine(NE)and dopamine(DA)were measured by Enzyme Linked Immunosorbent Assay(ELISA).Morphological changes were examined by optical microscopy.Results :(1)the general situation: the normal group of rats weight gain significantly,good appetite,hair luster,normal stool,eyes have God,mental state is good,free movement.The rest of the group after 10 days of modeling weight gain was not obvious,the amount of food intake and activity decreased,back curled up and so on,followed by squinting,coughing shortness of breath,sparse dull hair,hair removal,vertical hair,weight loss,food intake and activity significantly reduced,stool soft and partial thin,anal dirty cloudy,unresponsive,apathetic burnout.The rest of the group after 1 week of modeling weight gain was not obvious,the amount of food intake and activity decreased,back curled up and so on.During the treatment period,the response of the treatment group gradually improved after 1 week of treatment,the amount of food intake and activity increased compared with the previous,hair,stool,mental state than before have improved,while the general improvement of the model group is not obvious.(2)Serum IgA and IgG test results: IgA and IgG levels in the model group were lower than those in the normal group(P<0.01).The levels of IgA and IgG in Chaiwei group?Huangwei group and Jiawei group were higher(P<0.05)while contrasting with the model group,.There was no obvious diversification in IgA and IgG levels between the three groups(P>0.05).(3)Serum IgE test results: The IgE level of the model group was significantly increased while contrasting with the normal group(P<0.01).The level of IgE in Chaiwei group?Huangwei group and Jiawei group were significantly lower while(P<0.05),but it was no obvious difference in the level of Ig E between the three group(P>0.05).(4)Brain norepinephrine and dopamine test results:The concentration of DA in the brain of the model group was lower than that in the normal group(P<0.01).The DA concentrations in the Chaiwei group?Huangwei group and Jiawei group were higher when contracting with the model group(P<0.05 or P<0.01),and the DA concentration in the Chaiwei group was higher than that in the Huangwei group and Jiawei group(P<0.05),and no significant chance exists in the concentration of DA between the Huangwei group andthe Jiawei group(P>0.05).The concentration of NE in the model group was lower than that in the normal group,but there was no significant difference(P> 0.05).Compared with the model group,no significant difference exists in NE concentration between the three groups(P>0.05).(5)Pathology:In the model group,the bronchial mucosa epithelial cells adhered,a large number of inflammatory cell infiltration,pulmonary interstitial edema,wall congestion and expansion were obvious;gastric tissue showed atrophy of gastric mucosa,mucus increased,inflammatory cell infiltration;small intestine visible villous shedding,epithelial Cell atrophy,interstitial edema,after treatment was significantly restored,but the treatment group no significant difference.Conclusions:(1)Using the smoked,papain fog aerial injection can successfully replicate the rat model of spleen and spleen deficiency.(2)Lung and spleen deficiency model rats involved in digestion,immunity,nervous and other disorders,the brain monoamine neurotransmitters in the presence of metabolic disorders.(3)CWYG San,HWYG San and JWYG San are increased lung and spleen deficiency rats body weight,improve symptoms and signs;They can enhance the immune function of rats with deficiency of spleen and lung,increasing serum IgA?Ig G concentration,while reducing serum IgE concentration;They can improve lung and spleen deficiency in rats lung,bronchial,gastric and small intestinal tissue pathology,reduce the degree of damage and inflammation.They can make the brain monoamine neurotransmitter DA content increased,improve the state of neurological disorders.The effect of CWYG San on improvingthe DA in the brain of rats with deficiency of lung and spleen was better than that of HWYG San and JWYG San.The research indicates that Jianpi Yiqi?Jianpi Yangfei?and Shugan Jianpi Yangfei can enhance the lung and spleen of the body's immune function,and in improving the body's neurological disorders,Shugan Jianpi Yangfei(suppression of soil and soil enrichment method)is superior to the soil and the spleen and spleen-raising method.
