Objective: To evaluate the protective function of ethanol extract from Trillium tschonoskii Maxim(TTM)on CC l4-induced acute liver injury of rats.Methods: Totally 48 rats were randomly divided into 6 groups including control group,CCl4-induced acute liver injury model group,Dimethyl dicarboxylate biphenyl(DDB)treatment group,and TTM treatment at low,moderate and high dosage groups.A fter adaptive feeding for 7 days,30 rats except the control group were subjected to subcutaneous injection of 25% CCl4 at 5 m L/kg for the establishment of acute liver injury model.After model establishment,the rats from DDB group were administrated with DDB at the dose of 0.2 g/kg·d through lavage and the rats from TTM groups were administrated with TTM at various doses(0.1,0.5 and 1.0 g/kg·d)at the same volume through lavage,while the rats from control and model groups were administrated with saline at the same volume.Upon the treatments with DDB and TTM for 5 consecutive days,all rats were sacrificed for the collection of blood and livers.The liver index to body weight was calculated,and the levels of serum ALT and AST were determined.Meanwhile,the m RNA expression of TNF-? and IL-6 were evaluated with fluorescence quantitative real-time polymerase chain reaction(RT-PCR).Moreover,the sections of liver tissues were examined by HE staining and Tunel staining.Furthermore,the protein expression of Caspase-3,Bcl-2 and Bax was evaluated by western blot.Results: CCl4 could induce obvious acute liver injury and abnormal liver functions in rats with characteristics of hepatomegaly,increased liver indices,high levels of serum ALT and AST,up-regulated expression of inflammatory factors including TNF-? and IL-6,and activated apoptosis due to overexpressed Bax and Caspase-3.In contrast,DDB and TTM at various dosages could execute protective role in CC l4-induced acute liver injury in rats through reducing the levels of ALT and AST,rescuing hepatomegaly,down-regulating inflammatory factors and inhibiting apoptosis of liver cells.Moreover,the protective role of TTM in acute liver injury revealed an obvious dose-dependent manner.Conclusion: TTM has an obvious protective effect on acute liver injury in rats induced by CCl4 through inhibiting the apoptosis of liver cells.But it is not promote liver regeneration.
