Study On The Mechanism Of Ganwei Baihe Decoction Repair Model Mice's Gastric Mucosa Injury Caused By Chronic Stress And Helicobacter Pylori

Objective:To explore Impacts and action mechanism of Ganwei Baihe Decoction on mouse gastric mucosa HSP70,NF-kappa B,p-ERK,TFF1 protein expression in the model of mice with Helicobacter pylori infection and chronic stress.Methods:(1)60 BALB/c male mice were randomly divided into blank group,stress group,Hp group,double factors group,Ganwei Baihe Decoction group,ranitidine group,each group of 10 rats.(2)Blank group and Hp group were fed routinely,the other 4 groups were treated with unpredictable chronic stress method to prepare the mouse model,which lasted for 28days;When modeling 7 days,Given the Hp group,double factors group,Ganwei Baihe Decoction group,ranitidine group by intragastric administration of Hp bacteria liquid.When modeling 14 days,Ganwei Baihe Decoction group and ranitidine group were given corresponding drugs by gavage,the other 4 groups were given distilled water by gavage,all continuous gavage 14 days;(3)To test serum Hp antibody by Colloidal gold method;(4)To calculate the ulcer index and ulcer inhibition rate of mice in each group;(5)To observe the mice gastric mucosa pathological changes by HE stain;(6)To detect the mouse gastric mucosa of HSP70,NF-kappa B,p-ERK,TFF1 protein expression by immunohistochemical assay.Results:(1)Ganwei Baihe Decoction group Hp infected mice was significantly lower than that of the Hp group,double factors group and ranitidine group,the difference was statistically significant(P<0.01).(2)Double factors group gastric mucosal injury index increased compared with other groups,the difference was statistically significant(P<0.01).Compared with double factors group,the gastric mucosal injury index was decreased in Ganwei Baihe Decoction group and ranitidine group,the difference was statistically significant(P<0.01).And Ganwei Baihe Decoction group was lower than ranitidine group,the difference was statistically significant(P<0.01).(3)Compared with blank group,the HSP70,NF-kappa B,p-ERK,TFF1 of double factors group increased expression,the difference was statistically significant(P<0.01).Compared with double factors group,the HSP70,p-ERK,TFF1 of Ganwei Baihe Decoction group and ranitidine group increased expression,and the NF-kappa B decreased expression,the difference was statistically significant(P<0.01).The expression of HSP70,TFF1 and p-ERK protein in Ganwei Baihe Decoction group was higher than that in ranitidine group,and the expression of NF-kappa B protein was lower than that of ranitidine group,the difference was statistically significant(P<0.01).Conclusion:Ganwei Baihe Decoction can promote the expression of HSP70,TFF1 and p-ERK,promote gastric mucosa repair,enhance immunity,reduce NF-kappa B activity,reduce the gastric mucosal inflammation,so as to achieve the eradication of Hp,repair of gastric mucosal injury,promote ulcer healing.
