Effect Of Ganwei Baihe Decoction On HPA Axis MR/GR Balance In Chronic Stress Gastric Ulcer Model Mice

Objective: To explore the effect of Ganwei Baihe Decoction on HPA axis of chronic stress gastric ulcer model mice.Methods: 1.40 male BALB/c mice were randomly divided into control group,model group,Ganwei Baihe group,ranitidine group,the control group were given normal feeding,the other three groups can not be prepared in accordance with the mouse model of chronic stress predict method,continuous modeling for 28 days;14 days after modeling,give the corresponding drugs by gavage to the Ganwei Baihe Decoction group and ranitidine group,to the control group and the model group distilled water by gavage for 14 days,all the mice were observed;2.The general behavior change,and the use of open field test to observe the behavior change;3.Were calculated in each group,The ulcer index(UI)and the ulcer inhibition rate;4.Using HE to observe the pathological changes of gastric mucosa staining;5.The content of ELISA was used to detect serum GC,ACTH,CRH;6.Expression was detected by immunohistochemical method in mice hippocampus GR,MR protein.Results: 1.14 days after modeling,the model group,Ganwei Baihe Decoction group,ranitidine group were obviously abnormal behavior,open field test level score,vertical score,clean score and weight were significantly lower than the control group,the difference was statistically significant(P <0.01);Gavage for 14 days,The Ganwei Baihe Decoction group and ranitidine group mice to improve behavior,open field test level s score,vertical scores,clean score and body weight compared with the model group increased significantly,the difference was statistically significant(P <0.01),and the Ganwei Baihe Decoction group scores higher than ranitidine group,the difference was statistically significant(P <0.05);2.The Ganwei Baihe Decoction group,ranitidine group UI was significantly lower than that of model group,the difference was statistically significant(P <0.01),Ganwei Baihe Decoction group UI than ranitidine group,difference was statistically significant(P <0.05);Ganwei Baihe Decoction group,the ulcer inhibition rate was significantly higher than that of ranitidine group;3.To observe the Ganwei Baihe Decoction group and ranitidine group of mice gastric mucosa HE staining,only a small amount of erosion,erosion and scattered in the model group showed punctate ulcer;4.Compared with the ctrol group,Model group,Ganwei Baihe Decoction group,ranitidine group,serum GC,ACTH and CRH were significantly higher,the difference was statistically significant(P < 0.01);compared with the model group,Ganwei Baihe Decoction group,ranitidine group,serum GC,ACTH and CRH were significantly lower,the difference was statistically significant(P < 0.01);combined Ganwei Baihe Decoction group,serum GC,ACTH and CRH were lower than ranitidine group,the difference was statistically significant(P < 0.05);5.Compared with the control group,model group,Ganwei Baihe Decoction group,ranitidine group in hippocampus,the expression of GR protein decreased,MR protein expression increased significantly,the difference was statistically significant(P < 0.01);Compared with the model group,Ganwei Baihe Decoction group,ranitidine group GR expression increased,MR expression decreased obviously,the difference was statistically significant(P < 0.01);Ganwei Baihe Decoction group and ranitidine group comparison of GR expression increased,MR expression decreased,the difference was statistically significant(P <0.05).Conclusion: The mechanism of Ganwei Baihe Decoction in the treatment of chronic gastric ulcer of liver and stomach qi stagnation may be through the regulation of HPA axis,GC and CRH,reduce the secretion of ACTH,the hippocampus GR protein expression increased,MR protein expression was down regulated,change the state of chronic stress imbalance MR/GR relationship,reduce gastric mucosal inflammation.And then to repair the gastric mucosa,promote ulcer healing.
