post-traumatic stress disorder(PTSD)is a kind of disease because of the disaster,war,terrorist attack,abuse,such as traffic accident caused great pain or frightened,a tragedy caused by mental,physical and mental symptoms of persistent state or anxiety syndrome.In addition,hypothalamus-pituitary-adrenal axis(HPA axis)is an endocrine axis closely related to stress response.The hippocampus is the brain tissue upstream of the HPA axis.The hippocampal structure also plays an important role in learning and memory.Therefore,if the hippocampal structure or the structure associated with it is impaired,it leads to amnesia syndrome.Patients with PTSD had a sustained low cortisol response and a certain degree of hippocampal imaging and morphological changes,as well as significant enhancement of HPA axis negative feedback inhibition.Studies have shown that women with PTSD GR expression in hippocampus of drop degree is slightly higher than the male,the expression of hippocampal GR a large range than men,the role of MR in women is to be able to significantly reduce the incidence of PTSD.This research uses imitated PTSD rats model of a single continuous stimulation model single prolonged stress(SPS),by comparing the normal male and female rats and SPS behavioral changes of male and female rats and the change of the expression of hippocampal neurons MR and GR explore gender differences caused by PTSD morphological differences and molecular mechanism and MR,GR is involved in regulating PTSD gender differences of the disease.The hippocampus thought to be an important structure involved in learning and memory,located in the lower base of the lateral ventricle,is an important part of the limbic system.The amygdala is an important subcortical nucleus in the limbic system.Methods: Single-Prolonged Stress(SPS)was used to construct animal model of PTSD.Behavioral methods were used to detect behavioral changes in rats.Morphology methods included the Morris Water Maze Test(MWM),the Plus Maze Test and the Open Field Maze Test.Morris water maze was used to detect the spatial learning and memory abilities of rats in each group.Elevated plus Maze Test and Open Field Maze Test were used to detect the changes of anxiety in each group.Select a Single continuous stimulation method to build PTSD animal model.Western blot method was used to detect the differences in expression of the hippocampus GR and MR protein in rats after SPS stress.The changes in the subcellular distribution and immunoreactivity expression in the hippocampal and amygdala neurons in the hippocampal and amygdala neurons before and after SPS stress were measured.Results: The results of western blotting detecting showed that both MR and GR induced by SPS were increased in hippocampus and amygdala,while MR and GR also corrected the changes.Open field experiment and overhead cross experiment testing results show that the SPS group of male rats and female rats compared with normal control group male and female rats of self-exploration ability lower,than the male and female rats of self-exploration ability is more apparent.Morris water maze experiment showed that female rats and male rats were severely impaired in spatial memory after SPS stress,and their spatial memory ability was significantly reduced compared with normal group.Immunofluorescence double standard shows female rat hippocampal neurons in the normal group,GR was concentrated distribution in nerve cells in the nucleus,and in the male rats,GR rendering diffuse distribution,are mainly distributed in the cytoplasm of nerve cells,a small amount of distributed in the nucleus.There was no significant difference in the distribution of MR in male and female rats,mainly in the cytoplasm of nerve cells.After the stimulation of SPS,the immunoreactivity of GR and MR in the hippocampal neurons of the male and female rats decreased significantly compared with that in the normal control group,and there were significant differences in the distribution of sub-cells.SPS group male and female rat hippocampus of GR and normal gay compared to control group rats appear more diffuse distribution,mainly distributed in the cytoplasm and protuberant,also the same as the expression of GR and MR,compared with normal control group gay rats,rendering more diffuse distribution of cells.Western blot method detection results show that the SPS group of male and female rats hippocampal GR,MR gay rats were significantly lower than normal control group,and the female rats GR and MR expression quantity decreased significantly compared with male rats.Conclusion: 1.There is a gender difference between male and female in normal rats,and the female rats’ autonomous exploration ability and spatial memory ability aresignificantly lower than that of males.After SPS,the independent exploration ability and spatial memory ability of male and female rats decreased significantly compared with normal group.2.Normal female rats in the hippocampus were mainly distributed in the nucleus in comparison with male rats.MR is distributed in cytoplasm without significant gender difference.Compared with the normal control group,the GRS and MR in the hippocampus of female male rats were distributed(mainly in cytoplasm),and the immunoreactivity and protein total showed a significant decrease.3.The behavioral differences between male and female rats induced by SPS stimulation may be related to the differences in the expression of GR in hippocampal neurons of male and female rats. | |
