The Effect Of Huoxue Tongluo Decoction On Expression Of DLL4? Jagged1 And VEGF In The Process Of SANFH Rabbit Model

ObjectivesTo approach the curative effect of traditional Chinese drug and the mechanism of action on steriod induced avascular necrosis of the femoral head (SANFH).Methods192 rabbits were randomly divided into model group A,model group B,model control group C,blank control group D,each 48,A, B, C group used the modeling method he xijing modeling.The A group was given traditional Chinese medicine feed from the beginning of the second week,C,D group were given the same amount of common feed.At the end of second,fifth,eighth week do the femoral head pathology calculate the ratio of empty lacuna,using in situ hybridization(ISH) of rabbit femoral head VEGFmRNA were determined. And using the method of enzyme-linked immunosorbent (Elisa) in rabbit serum BMP2 for the determination of real-time fluorescent quantitative PCR (rt-pcr) in rabbit serum protein Jagged 1 gene expression were determined.ResultsAt the end of eighth week, the concentration of VEGF in C group was significantly lower than that in D group (p< 0.01), A group VEGF concentration was higher than C group (p< 0.05), the concentration of VEGF in B group were higher than those of group C (p> 0.01); at the end of eighth week,the rabbit femoral head VEGFmRNA consistent with rabbit serum VEGF concentration change;D114 expression in A group,B group,C group and D group showed increasing trend. At the end of eighth week,the expression of D114 in A group and B group was lower than that in C group (p< 0.05),but The growth rate of A group was greater than that of C group. At the end of fifth, eighth week,the expression of Jagged 1 in C group was higher than D group (p< 0.01),At the end of fifth week, A group was higher than C group (p<0.05), At the end of eighth week, each group weren't significantly (P>0.05), but Jaggedl expression in A group,B group,C group and D group showed increasing trend; At the end of eighth week,C group of empty lacuna rate was significantly higher than group D(p<0.01),A group,B group of empty of lacuna rate were significantly lower than C group (p< 0.01)ConclusionShow that for the prevention of the necrosis of the femoral head of TCM for Huoxue Tongluo Decoction treatment function exactly;2> May be of TCM for Huoxue Tongluo Decoction by down-regulated D114 and Jaggedl and maintain the concentration of VEGF femoral head, jointly promote bone repair and treatment effect on angiogenesis.
