Objective:To explore the expression of ELTD1,VEGF and DLL4 in gastric adenocarcinoma and their revelance,and clarify the relationship between ELTD1,VEGF and DLL4 with tumor angiogenesis,tumor cell proliferation and hypoxia,in order to provide a better biological indicator to evaluate the biological behavior of gastric cancer,and provide new targets for the treatment of GC and theoretical basis.Methods:Immunohistochemical staining was used to detect the expression of ELTD1,DLL4,VEGF,CD34,and CA9 in 43 cases of gastric adenocarcinoma tissues,5 cases of paracancerous tissues,4 cases of chronic gastritis tissues,and 7 cases of healthy human gastric tissues.Meanwhile,the clinicopathological parameters and expression levels of Ki-67 positive cells were recorded.SPSS23.0 statistical software was used for statistical analysis.Result:(1)There was no correlation between ELTD1 expression in gastric cancer tissues and the clinical stage,lymph node metastasis,depth of tumor invasion and histological differentiation of gastric cancer(p>0.05).(2)The expression of VEGF was significantly correlated with the clinical stage,lymph node metastasis,depth of tumor invasion and the degree of tissue differentiation in gastric cancer(p<0.05).(3)The expression of DLL4 was correlated with the clinical stage,lymph node metastasis,and depth of tumor invasion(p<0.05),but not with the degree of differentiation of gastric cancer(p>0.05).(4)There was no correlation between the expression of ELTD1 in gastric carcinoma and the expression of VEGF,DLL4,CD34,CA9 and Ki-67(p>0.05).(5)The expression level of VEGF in cancer tissues was positively correlated with the expression level of CD34(p<0.01,r_s>0),and negatively correlated with the expression level of CA9(p=0.01,r_s<0),but it had no significant correlation with Ki-67(p>0.05).(6)The expression level of DLL4 in cancer tissues was positively correlated with the expression level of CD34 protein(p<0.01,r_s>0),and negatively correlated with the expression level of CA9(p<0.01,r_s<0),but not correlated with Ki-67(p>0.05).(7)There was no significant difference in the expression of ELTD1 in gastric cancer tissue,paracancerous tissue,chronic gastritis tissue,normal gastric mucosa tissue and other different tissues(p>0.05).(8)There were significant differences in the expression of VEGF protein in 4 different tissues,such as gastric cancer,para cancer,chronic gastritis,and normal gastric mucosa(p<0.05).The comparison between the expression of the protein in gastric cancer tissues and other several tissues found that there were significant differences in the expression of VEGF between gastric cancer tissues and adjacent tissues,chronic gastritis tissues and normal gastric mucosa tissues(p<0.05).(9)There were significant differences in the expression of VEGF protein in 4 different tissues,such as gastric cancer,para cancer,chronic gastritis,and normal gastric mucosa(p<0.05).The comparison between the expression of the protein in gastric cancer tissues and other several tissues found that there were significant differences in the expression of VEGF between gastric cancer tissues and chronic gastritis tissues and normal gastric mucosa tissues(p<0.05),but there was no difference with paracancerous tissues(p>0.05).Conclusion:(1)The expression of ELTD1 in gastric cancer was not related to microvessel density,the degree of hypoxia,the proliferation,the occurrence and development of gastric cancer.This study failed to demonstrate that ELTD1 expression is regulated by VEGF and DLL4.The expression and role of ELTD1 in gastric cancer remains to be further studied.(2)The expression of both DLL4 and VEGF proteins were related to the occurrence and development of gastric malignancies.The expression of DLL4 and VEGF were negatively correlated with the degree of tissue hypoxia and positively correlated with the microvessel density of gastric cancer.The results showed that VEGF and DLL4 are important regulators in the development of gastric cancer.
