The extraction and contrastive study on the chemical composition of Salvia scapiformis Hance from Mao Ping,Wu Dong,Que Niao,Ge Tou,Gao Yan,Jiao Yao,Fang Xiang,Dou Zhai,Jiao La,Ping Xiang from Leigong Mountain in Guizhou Province,have been studied in this dissertation.Firstly,to study the difference on extraction methods of the S.Scapiformis,and to explore the best extraction conditions,the different proportions of methanol-water,ethanol-water solution,ethyl acetate and petroleum ether are used as soxhlet extraction and ultrasonic extraction solvent to ectract S.Scapiformis.The absorbance is measured by ultraviolet-visible spectrometer,which is used to estimate the difference concentration of the different extraction conditions.SPSS software is used to design orthogonal conditions,and to optimize extraction methods.The best extraction methods in different solvents are as follows: forty percent methanol-water system 25 ml to extract 2 h with ultrasonic,and the ?max in methanol-water system is286 nm,fifty-five percent ethanol-water system 15 ml to extract 1.7 h,and the ?max in ethanol-water system is 286 nm,ethyl acetate 20 ml to extract 1.5 h,and the ?max in ethyl acetateis is 270 nm,petroleum ether 10 ml to extract 1 h,and the ?max in petroleum ether is 214 nm.Secondly,using the best in methanol-water solution extraction to extract S.Scapiformis from different regions.Analysis the difference of main component by using IR fingerprint,it was found that the specific different components from different regions is small,so the main chemical component are the same basically.And then,analysizing the difference of main components through HPLC,we found that retention time(Rt)in 60.04 min(peak 8)and 61.31 min(peak 9)were the highest levels substance in ten regions,which were the main constitutes of methanol-water extraction.Peak 1(4.97 min),the highest content from Jiao La(6.26%),the lowest content(2.80%)from Fang Xiang.As peak 2,peak 3 and peak 4,the content of this three compounds have little difference.Peak 5(41.15 min),the highest content of S.Scapiformis is 8.77% from Jiao Yao,the lowest content of S.Scapiformis is 4.79%IV from Ping Xiang.Rt in 43.97 min(peak 6),the highest content of S.Scapiformis is4.62% from Que Niao,the lowest content of S.Scapiformis is 3.90% from Jiao La.Rt in 46.87 min(peak 7),the highest content of S.Scapiformis is 6.35% from Ping Xiang,the lowest content of S.Scapiformis is 1.46% from Jiao La.Rt in 61.31 min(peak9),the highest content of S.Scapiformis is 21.27% from Ping Xiang,the lowest content of S.Scapiformis is 14.83% from Fang Xiang.Finally,In order to extract liposoluble substance,petroleum ether 10 ml to extract S.Scapiformis 1 h with ultrasonic from the ten regions.Analysizing the difference of components by using GC-MS,9,12-Octadecadienoic acid was found to take the most share and n-Hexadecanoic acid take the second place with9,12-Octadecadienoic acid ahead.The content of Caryophyllene oxide from the ten regions are lower than 2% with the highest content of 1.42% from Que Niao and the lowest content of 0.32% from Mao Ping.Phenol.2,4-bis(1,1-dimethylethyl)was not found in Mao Ping,the highest content is 5.02% from Ge Tou.the highest content of Cedrol is 2.73% from Que Niao,the lowest content is 0.43% from Mao Ping.The content of 2,6,6-trimethyl-bicyclo(3.1.1)heptan are different from different regions with the highest content of S.Scapiformis is 8.95% from Mao Ping,19 times comparing to the lowest of 0.47% from Fang Xiang.The highest content of Methyl trans-9-Octadecenoate is 10.20% from Que Niao,while the lowest content is only1.91% from Mao Ping.The content of Phytol reaches as highest as 11.98% from Gao Yan and the lowest content of S.Scapiformis is 2.53% from Mao Ping.The content of9,12-Octadecadieneoic acid is higher than other compounds in all the ten samples with the highest of 39.50% from Fang Xiang.The content of Caryophyllene oxide and Cedrol was the highest in the area of Que Niao.In Mao Ping,the content of2,6,6-trimethyl-bicyclo(3.1.1)heptan was the highest,but the Caryophyllene oxide,Cedrol,Phenol,1-Tetradecen,2,4-bis(1,1-dimethylethyl)and 9-Octadecenoic acid,methyl ester was the lowest.The content of 2,4-bis(1,1-dimethylethyl)was the highest in Ge Tou.In Fang Xiang,the content of n-Hexadecanoic acid and9,12-Octadecadienoic acid was the highest and 2,6,6-trimethyl-bicyclo(3.1.1)heptan and 3-hydroxyflavone was the lowest.The content of Phytol was the highest in thearea of Gao Yan.The fingerprint chromatography can be used to study the chemical constituents of the extraction of S.Scapiformis.As shown above,not only can we judge the origin of S.Scapiformis but also be able to evaluate the quality of S.Scapiformis.
