Salvia yunnanensis C.H. Wright is a Chinese herbal medieine which is widely used in China. The Tibetan folk drug "DianDanShen" is the dried root of Salvia yunnanensis C.H. Wright, which is widely distributed in the west of China and has a similar pharmacological activity with Salvia Miltiorrhiza. On this basis, we Launched a research on the chemical components of Salvia yunnanensis C.H. Wright under the direct of an antiangiogenic activity. The main results are summarized below:1. Screening the activities of the extract of Salvia yunnanensis C.H. Wright with the chorioallantoic membrane angiogenesis model:Test the activity of the parts of the extract of the underground part of S. yunnanensis which separated with gradient column chromatography by the Petroleum ether-EtOAc9:1,8:2,2:1,1:1,1:2and the parts of the extract of the aerial part of S. yunnanensis which separated with gradient column chromatography by the Petroleum ether-EtOAc9:1,7:3,1:1. Find out that the parts of9:1,1:1of the underground part extract has significant antiangiogenic activity, the parts of8:2,1:2of the underground part extract showed significant pro-angiogenic activity,2:1part of the underground part extract did not show angiogenesis activity, the9:1part of the aerial part extract showed little antiangiogenic activity,1:1part of the aerial part extract showed significant antiangiogenic activity,7:3part of the aerial part extract showed significant Pro-angiogenic activity. Combined the experimental results, we chose to study the chemical components of the parts of9:1,8:2of the underground part and the parts of9:1,1:1of the aerial part.2. The separation of the underground and aerial parts:22compounds had been isolated from the9:1,8:2parts of the underground part such as tanshinone Ⅰ, tanshinlactone, tanshinone ⅡA, Danshenxinkun B, Methylenetanshinquinone,1,2-dihydrotanshinone Ⅰ,5,6-dehydrosugiol, sugiol,6α-hydroxysugiol, compound10, compound11, Crypotanshinone, Dihydrotanshinone, compound14and8compounds had been isolated from the9:1,1:1parts of the aerial part (stems and leaves) such as (+) Spathulenol,3β-Hydroxy-12-ursene, β-Amyrin, tanshinlactone,5,7,4’-Trihydroxy flavanone,2α,3β-dihydroxyursa-12-en-28-oic acid, Ursolic acid, 3-Oxo-12-ursen-28-oic acid.3. Screening the antiangiogenic activity of compounds isolated from Salvia yunnanensis C.H. Wright with the chorioallantoic membrane angiogenesis model: Test tanshinone Ⅰ(Y-1), tanshinlactone(Y-2), tanshinone ⅡA(Y-3), Danshenxinkun B(Y-4),5,6-dehydrosugiol(Y-5),6α-hydroxysugiol(Y-6), sugiol(Y-7), Crypotanshinone(Y-8), Dihydrotanshinone(Y-9), compound14(Y-10),(+)Spathulenol(Y-11),3β-Hydroxy-12-ursene(Y-12), β-Amyrin(Y-13),2α,3β-dihydroxyursa-12-en-28-oic acid(Y-14), and Ursolic acid (Y-15). Find out7compounds which have the antiangiogenic activity, such as tanshinlactone(Y-2), tanshinone IIA (Y-3),6a-hydroxysugiol (Y-6), sugiol (Y-7), Crypotanshinone (Y-8), compound14(Y-10),2α,3β-dihydroxyursa-12-en-28-oic acid(Y-14). The activity sequence of the compounds is Y-14> Y-2> Y-3≈Y-6≈Y-7> Y-10> Y-8.4. Analysis of chemical components in4Salvia Plants by High Performance Liquid Chromatography with Column Switching Technique:With using HPLC with the column-switching technique, we achieved the separation and analysis7active components (Danshensu, Protocatechuic aldehyde, Protocatechuic acid, Caffeic acid, Cryptotanshinone, Tanshinone Ⅰ and Tanshinone ⅡA) of the hydrophilic and hydrophobic components in4Salvia plants. The content sequence of4hydrophilic components is S. miltiorrhiza> S. yunnanensis C. H. Wright> S. przewalskii Maxim> S.castanea Diels, and the content of hydrophobic components in S. przewalskii Maxim and S.castanea Diels is much higher than in S. miltiorrhiza.Conclusion:we have isolated22compounds from S.yunnanensis C. H. Wright,7of which have the antiangiogenic activity. Compared with S. miltiorrhiza, Salvia plants have the similar chemical components,but the content of which is different in the use Salvia plants should be used selectively.
