The extraction and separation of the chemical composition and the HPLC fingerprint of Eomecon Chionantha Hance, have been studied in this dissertation.1.Studies on the extraction and separation: Air-dried and powdered E.Chionantha Hance(12 kg) was extracted with 75% ethanol every week for four times and pH=1 Hydrochloric acid. After removing the solvents under reduced pressure, the residue was obtained. The residue was suspended in water and then extracted successively with petroleum ether, chloroform, ethyl acetate and n-butanol to corresponding give four fractions. Two compounds were isolated and purified from petroleum ether part by column chromatography and recrystallization. β-Amyrin and Chelerythridimerine were identified by spectral methods ultraviolet(UV), infrared(IR) and 1H nuclear magnetic resonance(1H-NMR).2. Study on HPLC fingerprint: In order to obtain the best extraction conditions for the tested samples, cold soak extraction, ultrasonic extraction, soxhlet extraction and heating reflux extraction were operated to determine the appropriate solvent, the amount of solvent, extraction time and the ratio of material to liquid. According to the peak area in HPLC, the fingerprint chromatogram was researched. The results show that: 1. the separation degrees of chromatographic peaks by soxhlet extraction were lower than the other three methods and it contained a amount of small-content peaks, hence the separation degrees of cold soak extraction, ultrasonic extraction and heating reflux extraction were relatively higher compared with soxhlet extraction. 2.Cold soak extraction, ultrasonic extraction, soxhlet extraction and heating reflux extraction included many chromatographic peaks of HPLC finger-print, but the retention time in 13.276 min(peak 6) and 13.911 min(peak 7) indicated high level substance which were the main constitutes of E. Chionantha Hance under 320 nm. The percentage of peak 6 and 7 was 29.09% and 60.45%, respectively, with a total of 89.54% by the soxhlet extraction. Providing basic data for its’ quality control and component analysis, the HPLC fingerprint chromatogram can be used to study the chemical constituents of the extraction of E. Chionantha Hance.
