Background: To investigate the mechanism and active components is currently recognized for Traditional C hinese medicine(TCM) modernization. The efficacy of TCM frankincense are "Huo Xue, Xing Q i?Sheng Ji?Jie Du?Xiao Zhong?Zhi Tong". Frankincense can be used to treat a variety of inflammatory disorders, such as osteoarthritis and rheumatoid arthritis. Besides, it has been demonstrated that frankincense can be used as gastroprotective drugs and promote angiogenesis and wound healing. However, the mechanism and active components were still unknown. C linical symptoms of gastric ulcer is characterised with chronic recurrent epigastric pain, which is also called "gastric abscess". "Shu Gan" and "Li Q i" are the key therapeutic approaches in TCM therapy. Gastric ulcer is characterised with releasing of ROS and inflammatory cytokines, which is called "poison" in TCM. This aggravates ulcer formation and puts of the ulcer healing. Herbal Codification saies Frankincense belongs to "liver" and "spleen" in meridian theory. It could be used for ulcer treatment. O ur laboratory previously demonstrated that frankincense ameliorates cerebral ischemia-reperfusion oxidative stress injury through mediates Nrf2/HO-1, and improves blood stasis by e NOS activation. Once its gastroprotective and heal-promoting components and mechanism were proved. Frankincense might has the potential to be a better tolerated anti-inflamation drugs.Objective: To investigate the gastroprotective components and mechanism of Frankincense based on the TCM mechanism of "Jie Du" and "Sheng Ji".Method:(1)To investigate the gastroprotective effects of Frankincense extractives and find out the bioactive components of Frankincense in ethanol, indomethacin and thermal stress induced gastric ulcer in rats.(2) To assess the gastroprotective and anti-oxidative effects of ?-BA by measuring p H, GWM, PGE-2, MDA, SOD and C AT activity and the amount of nitric oxide(NO); and investigate the role of Nrf2/HO-1 in ethanol- induced gastric injury in rats.(3)To investigate the angiogenic and heal-promoting effects of ?-BA with indomethacin induced gastric ulcer in rats and by immunofluorescent staining?western blot?wound healing?transwell and tube formation assessment, and explain the "Sheng Ji" through VEGF/MMP-2 mediated signal.Resulsts:(1)The gastroprotective effects of Frankincense extractives have been verified and ?-BA might be crucial for gastroprotection.(2) ?-BA reduced injuries associated with the administration of ethanol, gastric juice acidity and the formation of MDA and increased CAT activity and SOD activity and the level of NO and PGE-2 in a dose-depended manner, and Nrf2/HO-1 pathway might be critical in ?-BA‘s gastroprotective effects.(3)?-BA could protect gastric ulcer formation through prevention of microcirculatory damage and blocking collagenolysis or promoting neovascularization and collagenization by recruiting VEGF and MMP-2.Conclusion: ?-BA could portect gastric ulceration through anti-oxidative effects and prevention of microcirculatory damage and blocking collagenolysis or promoting neovascularization and collagenization. The Nrf2/HO-1 pathway and VEGF/MMP-2 mediated signal plays a crucial role in gastroprotection and promotion of angiogenesis and wound healing.
