Objective To detect the expression levels of Nanog and PSCA in patients,tumor and adjacent to normal tissues of gastric cancer, and explore the relationship between stem cell marker and occurrence, progression, metastasis and prognosis of gastric cancer.Methods: The specimens were taken from October 2011 to June 2013 in the affiliated hospital of Inner Mongolia medical university of general surgery and surgical oncology, and gastric cancer surgical resection specimens of 100 cases by histologically confirmed. The expression of Nanog and PSCA in gastric cancer and corresponding adjacent normal tissues were examined by real-time PCR. And the correlational analyses was conducted to assess the relationship between Nanog and PSCA expression in gastric cancer tissue and clinicopathological factors. Results: Real-time PCR results showed that the relative expression level of Nanog in gastric cancer tissues was singnificantly higher than in adjacent to normal gastric tissues(P<0.05), the relative expression level of PSCA was below normal tissues adjacent to carcinoma(P<0.05). the Nanog and PSCA expression was associated with the differentiation status of gastric cancer. The poorly differentiated and undifferentiated group of Nanog was higher than moderately differentiated and well differentiated group(P<0.05), The poorly differentiated and undifferentiated group of PSCA were lower than moderately differentiated and well differentiated group(P<0.05), but no correlation were found between the expression and other clinicopathological parameters e.g. age, gender,tumor size, TNM stage, depth of invasion, and lymph node metasis. Conclusions: the Nanog and PSCA expression were correlated with the genesis and differentiation status of gastric cancer, can be used as new diagnosis of gastric cancer stem cell markers.
