Objective:Runt-related transcription factor3gene (RUNX3) and PSCA gene have t important relations with gastric cancer. Many researches have confirmed that rs2976392GG,, rs760805AA, rs2294008TT and rs2976392AA can increase the risk of gastric cancer. The purpose of the study was to investigate the relationships between the polymorphism of the RUNX3gene and the PSCA gene in gastric cancer.Methods:Both the RUNX3gene rs2236852and rs760805and the PSCA gene rs2294008and rs2976392genotypes were determined by PCR-based DNA sequence measuring analysis and direct DNA sequencing in310incident cases with gastric cancer and327controls recruited in Shandong Qingdao.Results:As to RUNX3gene rs2236852, the frequency of AA genotype was13.5%in gastric cancer patients and24.8%in normal controls, and the corresponding percentages for CG and AG genotypes were60.0%and44.6%, and26.5%and30.6%, As to rs760805,The frequency of TT genotype was15.16%in gastric cancer patients and20.49%in normal controls, and the corresponding percentages for AT and AA genotypes were48.4%and36.5%, and52.6%and26.9%, respectively. Compared to TT genotype, AT and AA genotypes were associated with an increased risk of gastric cancer (OR=1.24,95%CI=0.81-1.92; OR=1.83,95%GI=1.15-2.92). In rs2294008, the frequency of CC genotype was43.40%in gastric cancer patients and53.70%in normal controls, and the corresponding percent-ages for CT and TT genotypes were47.4%and9.2%, and39.0%and7.3%, respectively. Compared to CC genotype, CT and TT genotypes were associated with an increased risk of gastric cancer (OR=1.50,95%CI1.01-2.23;OR=1.55;95%CI1.77-3.15). In rs2976292, The frequency of GG genotype was43.23%in gastric cancer patients and50.95%in normal controls, and the corresponding percent-ages for GA and AA genotypes were41.9%and14.8%, and41.4%and7.6%, respectively. Compared to GGgenotype, GA and AA genotypes were associated with an increased risk of gastric cancer (OR:1.19,95%CI0.77-1.86) and (OR:2.30,95%CI1.13-4.66)。 Conclusion:The rs760805and rs2286352polymorphism of the RUNX3gene and the rs2976392and rs2294008of the PSCA gene are associated with gastric cancer.
