Objective:To study the expression and significance of FHIT protein,and survivin protein in endometrial carcinoma; to analyze the relationship between FHIT protein, survivin protein and the clinicopathological parameters of endometrial carcinoma.Method:Using the immunohistochemical S-P method, to detect expression of FHIT protein and Survivin protein in several biological tissue samples, which contain 20 cases of normal endometrium, 20 cases of simple endometrial hyperplasia, 20 cases of atypical endometrial hyperplasia and 60 cases of endometrial carcinoma(including 50 cases of endometrial adenocarcinoma, 6 cases of uterine serous carcinoma, 4 cases of clear cell carcinoma).Results :(1) Both FHIT protein and Survivin protein were expressed in the cytoplasm or nucleus.(2) The positive expression intensity of FHIT protein in endometrial carcinoma tissues was significantly lower than in atypical endometrial hyperplasia, simple endometrial hyperplasia and normal endometrium(Hc = 19.048, P<0.001). In endometrial carcinoma tissues, the degree of positive expression intensity of FHIT protein was related to tumor differentiation grade, clinical staging, depth of myometrial invasion and lymph node metastasis(Hc = 18.435-24.443, P <0.01), but had no relation to patient age, whether menopause or not, gross type, histological type(Hc =0.494-1.878, P> 0.05).(3) The degree of positive expression of Survivin protein in endometrial carcinoma was significantly higher than in atypical endometrial hyperplasia,simple endometrial hyperplasia and normal endometrium(Hc = 28.241, P <0.001). In endometrial carcinoma tissues, the degree of positive expression intensity of Survivin protein was related to tumor differentiation grade, clinical staging, depth of myometrial invasion and lymph node metastasis(Hc = 11.114-41.560, P <0.01), but had no relation to patient age, whether menopause or not, gross type, histological type(Hc = 0.174-0.438,P> 0.05).(4) In endometrial carcinoma tissues, expression intensity of FHIT and Survivin proteins through correlation analysis showed that both were highly significant negative correlation(r =-0.336, P <0.01).Conclusion:(1) Reduction or deletion of FHIT protein expression may be involved in the process of endometrial carcinoma, and has some connection with the development and invasion, metastasis of endometrial carcinoma.(2) Enhancement of Survivin protein expression is related to the development and invasion, metastasis ofendometrial carcinoma. It may have some significance for the early diagnosis of endometrial carcinoma.(3) Survivin protein and FHIT protein expression was negatively correlated, suggesting that antagonism plays in the occurrence and development of endometrial carcinoma.
