Objective To investigate the expression of Fhit and Survivin in human bladder transition cell carcinoma and vesicalleukoplakia. To examine the relationship between the Fhit expression and the Survivin expression in bladder carcinoma.Methods Immunohistochemical staining of Fhit and Survivin were performed by the standard streptavidin-peroxidate(SP) technique for the paraffin-embeded sections of 26 bladder transition cell carcinoma and 6 vesicalleukoplakia and 6 normal tissue. The relationship between expression of Fhit with the cliniopathplogic parameters, prognosis and the expression of Survivin was analyzed, respectively.Results 1. The positive rate of Fhit were 46.15% in BTCC, 100% in vesicallleukolakia and normal tissue. Respectively, the positive rate in BTCC of Fhit was lower than those in the vesicallleukolakia and normal tissue (P<0.05).2. The positive rate of Survivin protein were 69.51%(57/82), 30%(12/40)and 15%(3/29)in turn. The positive rate in BTCC of Survivin was higher than in the normal tissue (P<0.05).3. Loss of function of both tumor genes showed a positive correlations (P<0.05).Conclusions De-activation of Fhit and up-regulation Survivin expression might play synergefic roles in tumor genesis and development of BTCC. And it may be an early event in BTCC. And also might be helpful to making a diagnosis of or screening for bladder carcinoma. Survivin might involve a bad prognosis.
