In recent years, due to the environmental degradation, the incidence of tumor increased. Now most cases use comprehensive surgical operation, but the cure rate of cancer is still low. In China, traditional Chinese medicines have a long history, and it turns to be good in terms of practice. The clinical observation results showed that compared with radiotherapy and chemotherapy, Chinese medicines have obvious advantages in the aspects of tumor cell metastasis and recurrence.Chinese materias medica is rich from plant resources. Screening anti-tumor active substances traditional Chinese medicas has become a new hot field of researchers in recent years.Manchurian walnut(Juglans mandshurica Maxim.) belongs to Juglandaceae plant. Its growth environment is very widespread, mainly concentrated in the northeast and north China. It is a very important medicinal plant, and its seed, fruit and bark can be used as medicines which have anti-tumor, anti-oxidation, anti-bacterial, antiinflammatory, analgesic and other effects. As the incidence of tumor growth in recent years, the manchurian walnut get more and more attention.In this paper QIU-PI(Manchurian walnut bark) were researched. 8 compounds were obtained from n-butanol extract of QIU-PI by using macroporous resin, silica gel, Sephadex LH-20 column chromatography method and preparative high performance liquid chromatography and other modern separation technology. Using HPLC-MS, high resolution mass spectrum, nuclear magnetic resonance(NMR) spectrum methods, to identify the structure of these compounds are: naringenin(1), kaempferol(2), luteolin(3), oleanolic acid(4), protocatechuic acid(5), catechin(6), gallic acid(7) and hesperetin(8). Compounds(4),(5),(6) were found from QIU-PI for the first time.Then, under the guidance of the anti-tumor efficacy, the experiments in vitro using human esophageal cancer cell(EC-109) and human hepatoma cell(HepG-2) were conducted on QIU-PI extract ingredients, which discovered naringenin(1), kaempferol(2), luteolin(3), protocatechuic acid(5), catechin(6) and hesperetin(8) had obvious inhibitory effect on EC-109 cell and HepG-2 cell. According to the results calculated the IC50 value for EC-109 cell are: 0.38 mg/m L, 0.88 mg/m L, 0.38 mg/mL, 0.40 mg/m L, 0.81 mg/mL, 0.48 mg/m L, the IC50 value for HepG-2 cell are: 0.27 mg/mL, 0.40 mg/mL, 0.28 mg/mL, 0.33 mg/mL, 0.84 mg/m L, 0.27 mg/Ml.This study on anti-tumor activity for reference screening six active compounds from the n-butanol extract of QIU-PI exhibited significant anti-tumor activity, and 6 compounds from the n-butanol extract of QIU-PI displays excellent anti-tumor activity in vitro. In addition, this study choose human esophageal cancer cell line and human hepatocellular carcinoma cell line as the research object. The experimental result provides reference for further research of QIU-PI against esophageal cancer and liver cancer follow-up study.
