Manchurian Walnut Extract To Inhibit The Mechanism Of Tumor Cells

The ethanol extract of Juglans mandshurica leaves was extracted by petroleum ether, chloroform, and acetyl acetate in turn, followed by silica chromatograph. The red fraction in column shows the anti-tumor activity. Combining with the effect of cell cycle, proliferation, apoptosis, the anti-tumor mechanism of extract was discussed, at the same time, a screening system of anti-tumor was also built in this study.After treated the Hela, HLF, K562 cells with the extract for 24 h, MTT method was adopted to get the survival change, the LDso of Hela, HLF, K562 cells is 406.2, 154.5, 918.7ug/mL respectively. This demonstrated that the extract has distinct inhibiting effect on tumor, but no influence on normal cells. Furthermore, after treated with the extract, the telomerase activity was decreased, which showed that the extract maybe an inhibitor of telomerase.With the morphologic observation, agarose gel electrophoresis and flow cytometer, the apoptosis effect of Hela, K562 induced by the extract was confirmed. It showed that the extract has obvious influence on the cell cycle.These results show that this extract could inhibit the growth of tumor cells by influencing the cell cycle, inhibiting telomerase, inducing cell apoptosis, which can serve as the basis for developing the low-toxicity, high-performance anti-tumor drug.
