Objective To investigate the inhibitory effect of Ethyl acetate extracts of Impatiens Balsamina L. on prostate cancer cell line PC-3, RV-1, LNCaP.Methods Impatiens Balsamina L. was extracted and separated to get water extracts,ethanol extracts, oil ether extracts, ethyl acetate extracts and butanol extracts. Cell Counting Kit-8(CCK-8) assay was used to detect the inhibitory effect of Impatiens Balsamina L on human prostate cancer cell lines. Apoptosis rates and cell cycle distribution were detected by flow cytometry analysis. Transwell assay was performed to test the ability of migration. The expression of apoptosis-related proteins such as Bcl-2,Bax, cleaved-caspase-3, p-ERK, ERK, p-AKT, AKT were detected by western blot.Results Ethyl acetate extracts of Impatiens balsamina L had the strongest inhibitory effect on the growth of PC-3, RV1 and LNCa P cells, in comparison with the other extractsof Impatiens balsamina L. And EAEIB significantly inhibited the growth of PC-3, RV1 and LNCa P cells in a dose-dependent manner. After treated with different concentrations ethyl acetate extracts for 24 h, the percentage of G0/G1 phase increased significantly and apoptosis rate was significantly higher than the control group. After treated with different concentrations ethyl acetate extracts for 6h, the ability of migration of PC-3, RV1 were inhibited significantly. Western blot analysis showed that the expression level of Bcl-2,p-ERK, p-AKT were significantly decreased but the expression of Bax, cleavage of procaspase-3 were increased in comparison with the control group.Conclusions Impatiens Balsamina L inhibited the proliferation of human prostate cancer cell lines, ethyl acetate extracts have the strongest effect. Ethyl acetate extracts could inhibite cell proliferation, inhibit cell migration, cause G1 phase arrest and induce apoptosis through inhibition of the AKT and ERK pathways in human prostate cancer cells, indicating its potential as a therapeutic agent worthy of further development.
