Objective: The aim of this study was to examine whether single nucleotide polymorphisms(SNPs) of KCNMB1(?1 subunit of large-conductance Ca2+-activated potassium channel) and KCNJ11(inwardly rectifying potassium channel, subfamily J, member 11) are associated with essential hypertension(EH) in Xinjiang Kazak.Methods:(1) Whole blood genomic DNA extraction kit was used to extract genomic DNA in the blood of Kazakh from Xinjiang, including 267 essential hypertension patients( EH)and 259 normotensive group(NT), respectively.(2) Polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP) technique was applied to detect SNPs(rs2285676, rs5219 and rs11739136) of KCNJ11 and KCNMB1 gene.(3) The genotyping results were verified through the direct sequencing PCR products.(4) Hardy-Weinberg equilibrium was used to detect group representation of study population.(5) SPSS17.0statistical software package was applied to statistical processing and analysis for data from Excel. The unconditional logistic regression was used to estimated odds ratio(OR) and 95 %confidence interval(CI) for analyzing the risk factors related hypertension.Results:(1) Genotype frequency distribution of KCNMB1(rs11739136) and KCNJ11(rs2285676 and rs5219) polymorphism accorded with Hardy-Weinberg equilibrium in Kazak population from Xinjiang, which has demographic representation(P > 0.05).(2) Genotype CC,(CT+TT) and allele frequency(C, T) of the KCNJ11-rs2285676 were 4.12 %, 95.88 %,28.65 % and 71.35 % in EH group, respectively. Compared with NT group, there was no apparent difference between two groups(P > 0.05).(3) CC,(CT+TT) genotype and C, T allele frequency of KCNMB1-rs11739136 were 70.04 %, 29.96 %, 82.58 % and 17.42 % in EH group, respectively. Compared with NT group, there was no apparent difference between two groups(P > 0.05).(4) GG,(GA+AA) genotype and G, A allele frequency of KCNMB1-rs11739136 were 34.83 %, 65.17 %, 61.42 % and 38.58 % in EH group,respectively. Compared with NT group, there was significant difference between two groups(P < 0.05). Individual who carrying rs5219-GG genotype was 2.08 times than(GA +AA)suffered from EH(95 % CI: 1.40-3.08). Moreover, allele genotype carrying rs5219-G was1.68 times than A suffered from the risk of hypertension(95 % CI: 1.32-2.15).(5) The BMI(OR=1.09) and GG genotype(OR=2.15) of KCNJ11-rs5219 were relative factors affected hypertension in Xinjiang Kazak by unconditional logistic regression.Conclusions:(1) Our case-control study indicates that KCNMB1-rs11739136 and KCNJ11-rs2285676 maybe not associated with EH in Xinjiang Kazak.(2) KCNJ11-rs5219 gene maybe associated with EH. And carrying GG genotype may enhance the risk suffered from EH in Xinjiang Kazak.
