Objective: To detect the polymorphisms of 27 SNPs located on Cx gene in Xinjiang Kazak and Han Chinese, and to explore the correlation between SNPs and hypertension.Methods: ①The polymorphisms of connexin 40(connexin, Cx40) gene in 130 Kazakh patients and 150 healthy controls were detected with polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). ② Gene polymorphism of 1045 samples, including 259 cases of Kazakh patients with EH and 261 normal cases, and 260 cases of Han patients with EH and 265 normal cases,were determined by MALDI-TOF MS. ③SPSS17.0 statistical software package was used for statistical data processing and analysis, Hardy-Weinberg equilibrium test was used to judge the representative of the research objects.Results: ① In addition to 4 SNPs, the other SNPs conformed to Hardy Weinberg equilibrium(all P>0.05).②The frequencies of genotypes AA、AG、GG and alleles A and G of-44G>A in EH patients were 5.38 %、40 %、54.62 % and 25.38 %、74.62 %, respectively.No significantly difference was found between EH patients and controls(P>0.05). The frequencies of genotypes AA、AG、GG and alleles A and G of +71A>G in EH patients were 54.62 %、40 %、5.38 % and 74.62 %、25.38 %, respectively. No significantly difference was found between EH patients and controls(P>0.05). ③ rs1630310 SNP involved three genotypes in Kazak population, the frequencies of AA、AG、GG and A、G were(24.8 %、47.2 % 、28 % and 48.4 %、51.6 %) in Kazakh patients with EH, respectively, which were significantly different from those in Kazakh patients with EH(P=0.016,P=0.00).The frequencies of AA、AG 、GG and A、G were(24 %、52.8 % and 23.2 % and 50.4 %、49.6 %) in Han patients with EH, respectively, which were no significantly different from those in Han patients with EH(P=0.42,P=0.25). rs1925223 SNP involved three genotypes in Kazakh population, the frequencies of CC、CG and GG were 5.3 %、33.6 % and 61.2 % in Han patients with EH(P=0.37, P= 0.16)the frequencies of A and G were 22 % and 78 %, respectively, which were significantly different from those in Han patients with EH(P=0.048).The other genotype frequencies was no statistical differences between Han and Kazak EH patients and controls.Conclusion: Cx37 gene rs1630310 SNP maybe associated with EH. Cx43 gene rs1925223 SNP maybe associated with EH.Carrying rs1630310 A or rs1925223 G genes may have the protective effect on EH.
