Objective: To study the possible association between single nucleotide polymorphisms of SCN5A gene and bradycardia in Han Chinese people.Methods: A total of 8 unrelated hospitalized patients with bradycardia were enrolled. Genomic DNA was extracted from peripheral blood leukocytes. All 28 exons of SCN5A were amplified by polymerase chain reaction using 41 pairs primers. The PCR products were sequenced directly.Results:â‘ 7 SNPs of SCN5A gene were found. 3 of them were found in the coding region, including G281A, H558R and T5648C; and 4 of them were found in 3'-UTR,including A6365G, C6995T, 7204+7205TC>AT and +7698A.â‘¡The allele frequencies of G281A, H558R, T5648C, A6365G, C6995T, 7204+7205TC>AT and +7698A in Han Chinese people were 6.25%, 12.5%, 50%,62.5%, 31.25%, 68.75%, 62.5%, respectively.Conclusion: H558R and the four SNPs in 3'-UTR may attribute to bradycardia in different mechanisms.
