Automatic biochemical analyzer plays a critical role in clinical diagnosis and is mainly used to detect biochemistry indexes(e.g. hemoglobin, blood sugar, lipid and cholesterol) in human blood and urine. Reaction disc incubation system, which aims to control the temperature of the reaction mixture accurately and precisely, is one of the key technologies in automatic biochemical analyzer. Temperature control accuracy and heating rate of reagents cause massive effects on the accuracy of detection results, especially for reactions which involve enzymes.Four methodologies were used to manage reaction disc incubation system, which are air bath thermostatic methodology, direct contact heat methodology, water bath cycling and liquid thermostat cycling. While analyzing reaction disc's structure, heating pathway, and heating mode, we summarized their pros and cons in this article. A new methodology, hybrid bath thermostatic methodology(HBT), is firstly proposed concerning merits of direct contact heating and air bath thermostatic. Regarding heat transfer theory, we proceeded detailed analysis in HBT-mode reaction disc by heat conduction, heat convection, heat radiation, and gained corresponded heat flux of the three methods. We applied Runge-Kutta Method to solve differential equations and obtained the time needed for the thermostat to reach the preset temperature. By using ANSYS software to process finite element analysis(FEA) on HBT, we derived the level of influence of the vessels-contacted reaction disc and reagents' heating rate.This project also demonstrated a reaction disc incubation experimental platform; and developed a multi-channel thermal analysis temperature sensing module, to present an experiment-based demonstration in these two reaction-disc thermostatic controlling methodologies. Heat storage capacity of HBT-mode reaction disc is very good and the heat conduction rate of reaction disc is fast, so that it can enables the temperature of 300 ul reagent heated from 7? to 36? in 2.3 minutes, while resistance to temperature fluctuations is also strong and the temperature fluctuation of the reaction liquid is ±0.024?.
