Objective: This study focus on the differential expression of triose phosphate isomerase in human lung adenocarcinoma tissues and para-cancerous lung tissues as well as the differences in preoperative and postoperative serum samples.To analysis the correlation of the expression level of the TPI in serum and tissue.In order to research whether it can be used in the detection of serum sample. Therefore, providing a convenient and effective way to evaluate the diagnosis and prognosis of lung adenocarcinoma.Methods:(1)Human lung adenocarcinoma tissues and matched para-cancerous lung tissues of 44 cases from Hu-Nan Cancer Hospital of thoracic surgery department were acquired without radiotherapy and chemotherapy. Pathological diagnosis shows lung adenocarcinoma(Does not contain other cancerous tissue);(2)It contains 24 pulmonary benign disease patients without tumor history. The specimen was acquired from Hu-Nan Cancer Hospital of thoracic surgery department including bronchial pneumonia, pulmonary tuberculosis and inflammatory pseudotumor tissues.(3) Blood test was used in the two groups during the preoperative and postoperative period.Statistical analysis was used with mean difference: ELISA through Wilcoxon rank test, immunohistochemical results use rank sum test.Results: ELISA analysis result:(1)Comparing preoperative pulmonary benign disease group(A1) and postoperative pulmonary benign disease group(A2), use the Wilcoxon rank test,Z=0.517,Bilateral inspection(P = 0.605 > 0.05) shows no significant differences between groups;(2) Comparing preoperative lung adenocarcinoma group(B1) and postoperative lung adenocarcinoma group(B2),use the Wilcoxon rank test,Z=1.228,Bilateral inspection(P = 0.219 > 0.05) shows no significant differences between groups(3) Comparing pulmonary benign lesions and lung adenocarcinoma group, use the Wilcoxon rank test,Mann-Whitney U=430; Wilcoxon W=706; Z=0.517,Bilateral inspection(P = 0.787 > 0.05),P > 0.05, shows no significant differences between groups;(4)The comparison among two groups using the Wilcoxon rank test, Z=0.274,Bilateral inspection(P = 0.784 > 0.05), shows no significant differences between groups.Immunohistochemical analysis shows:(1)There are significant differences between two groups in the expression of TPI,p<0.01.The differences between the two groups were significant;(2)The expression of TPI shows no significant differences between normal lung tissue group and pulmonary benign disease tissue group, p>0.05;(3) The expression of TPI shows significant differences between normal lung tissue group and lung adenocarcinoma tissue group, P<0.01; The differences between the two groups were significant.(4) The expression of TPI shows significant differences between pulmonary benign disease tissue group and lung adenocarcinoma tissue group, P<0.01; The differences between the two groups were significant.Conclusion:(1) preoperative pulmonary benign lesions and lung adenocarcinoma group of difference of the serum levels of TPI density;(2) the preoperative and postoperative serum in lung benign lesions group TPI level was no significant change;(3) in lung adenocarcinoma group compared with preoperative serum after the TPI density declined, but without statistical significance;(4) the surgical treatment of preoperative postoperative serum concentration of TPI changes in lung benign lesions and lung adenocarcinoma group no significant difference.(5) the expression of TPI between pulmonary benign disease tissue and matched normal lung tissue have no obviously statistical difference,namely the TPI in lung adenocarcinoma tissue have a highly expression,TPI in pulmonary benign disease tissue and matched normal lung tissue have no expression or weekly positive expression, its hint that the TPI is a protein of differential expression.(6)Because of the expression of TPI has no significant difference between lung adenocarcinoma patient's serum and pulmonary benign disease patient's serum, The clinical significance of TPI enzyme in the blood test for lung adenocarcinoma has few significance.
