Research purposes Explore the influence on acute paraquat plasma concentrations of paraquat poisoning in SD rats with different gastric lavage time and different concentrations of bleaching earth,Provide a theoretical basis for the scientific development of clinical acute paraquat poisoning gastric lavage in patients with the optimal solutions.Research methods The random number table 48 rats were randomly divided into eight groups:A1 model group,A2 model group,B1 water group,B2 water group,C115% turbid liquid bleaching earth group,C230% turbid liquid bleaching earth group,D115% turbid liquid bleaching earth group,D230% turbid liquid bleaching earth group,N = 6,A1,B1,C1,D1 four groups administered 30 min after gastric lavage,A2,B2,C2,D2 four groups administered 60 min after gastric lavage,Take bleaching earth 30 g added in cup(Fuller's earth,CAS NO.:8031-18-3,purity-100 mesh particle size,the future provided by Shanghai Reagent Company),Add sterile distilled water 100 ml,formulated as a 30% bleaching earth suspension;The same method of preparation 15% bleaching earth suspension.Each rat was weighed before gavage on,For rats 150 mg / kg orally standard paraquat solution(Shanghai Pharmaceutical Co.,Ltd.Sinon,20% aqueous solution 20 ml / bottle),after administration 30 min,extraction within the rat orbital venous blood 2ml,placed anticoagulant tube,Centrifugal radius 20 cm,4000r / min,centrifugation 10 min,the serum stored at-80 ?,plasma paraquat concentrations by HPLC,Each group of rats paraquat concentration analysis of variance,the difference was not statistically significant explanation of acute paraquat poisoning rat model was successfully constructed.Take the same approach to detect plasma concentrations of paraquat in rats exposed to 1.5h,2.5h and 4.5h,The resulting data were entered SPSS double check entry method and factorial analysis,using mean ± standard deviation for each group of paraquat concentration in rat plasma descriptive analysis.Research result 1? plasma paraquat concentration at different time points:(1)After administered 30 min,plasma concentrations of paraquat A1 group,B1 group,C1 group,D1 group were respectively 52.97 ± 3.53,56.00 ± 4.53,54.60 ±3.94,53.35±9.70;A2 group,B2 group,C2 group,D2 group were respectively 54.60±5.28,56.27±4.88,55.82±4.52,54.78±4.50.(2)After administered 1.5h,plasma concentrations of paraquat A1 group,B1 group,C1 group,D1 group were respectively 251.22±6.92?210.15±8.49?161.48±5.83?132.60±7.37;A2 group,B2 group,C2 group,D2 group were respectively 283.97±7.89,240.67±6.05,195.68±6.27,167.22±3.62.(3)After administered 2.5h,plasma concentrations of paraquat A1 group,B1 group,C1 group,D1 group were respectively 266.15±7.78,216.68±8.39,173.73±4.66,143.73±7.63;A2 group,B2 group,C2 group,D2 group were respectively 291.38±9.74,246.82±9.72,201.32±8.52,166.90±7.32.(4)After administered 4.5h,plasma concentrations of paraquat A1 group,B1 group,C1 group,D1 group were respectively 228.00±8.07,162.73±5.24,123.42±6.58,84.62±4.44;A2 group,B2 group,C2 group,D2 group were respectively 240.85±5.01,183.72±5.30,142.37±5.10,111.72±5.12.2?Statistical analysis(1)The difference of plasma concentrations of paraquat in each experimental group was not statistically significant after 30 min rats exposed.Description that acute paraquat poisoning SD rat model was successfully constructed;(2)The Statistical Analysis results show that F=0.229,P=0.876 after exposure 1.5h,There is no interaction between the gastric washing concentration and gastric washing time;gastric washing concentration F=718.006,P<0.05 gastric washing time F=291.162,P<0.05,description of the differences between the two main effects were statistically significant,From descriptive statistics and analysis,Plasma concentrations of paraquat from high to low were respectively model group(267.59±18.51),shimizu group(225.41±17.42),15% bleaching earth suspension group(178.58±18.77),30% bleaching earth suspension group(149.91±18.91).(3)The Statistical Analysis results show that F=0.412,P=0.745 after exposure 2.5h,There is no interaction between the gastric washing concentration and gastric washing time;gastric washing concentration F=526.093,P<0.05 gastric washing time F=128.375,P<0.05,description of the differences between the two main effects were statistically significant,From descriptive statistics and analysis,Plasma concentrations of paraquat from high to low were respectively model group(278.77±15.63),shimizu group(231.75±17.96),15% bleaching earth suspension group(187.53±15.82),30% bleaching earth suspension group(155.32±14.04).(4)The Statistical Analysis results show that F=3.177,P=0.034 after exposure 4.5h,the interaction between the gastric washing concentration and gastric washing time shows that the difference was statistically significant;and the descriptive statistics and analysis shows that plasma concentrations of paraquat from high to low were respectively model group(234.43±9.72),shimizu group(173.23±12.05),15% bleaching earth suspension group(132.89±11.37),30% bleaching earth suspension group(98.17±14.87).Combining with the interaction contour map,30% bleaching earth suspension lavage after exposure 30 min were the best methods for acute paraquat poisoning,The two groups have synergistic action.(5)repeated measures analysis of variance on the plasma concentrations of paraquat at different time points in each group of rats longitudinal analysis,different concentrations of the gastric washing shows that the difference was statistically significant(F=815.17,P<0.05),different levels of gastric lavage time points difference was statistically significant(F=197.60,P<0.05),differences between the various time points were statistically significant(F=10359.40,P<0.05),differences between time points with gastric lavage concentration was statistically significant(F=371.368,P<0.05),A statistically significant difference between the different gastric lavage Times and Time(F=92.148,P<0.05);A combination of different time points gastric lavage MEASURE-1 estimated marginal trend means that the concentration of paraquat increased were delayed by gastric lavage in vivo plasma in rats,the effect shows best when the rats exposed to gastric lavage 30 min,with 30% of bleaching earth suspension.3? Lung tissue pathology results Model group(not gastric lavage group)shows the overall structure of severely abnormal in lung tissue,alveoli disappear,lung parenchyma,alveolar septa telangiectasia congestion,overall lung congestion;Water gastric lavage were observed in some alveolar lung tissue atrophy,alveolar septa thicken,part of the alveolar fusion into pulmonary bullae;15% bleaching earth suspension lavage group and 30% bleaching earth suspension lavage group shows that some abnormal exist in overall lung tissue structure,alveolar septa thicken with telangiectasia congestion,part of the alveolar fusion into pulmonary bullae,as well as peribronchial inflammatory cell infiltration.Conclusion Comparison between the water group,15% of bleaching earth suspension group,30% of bleaching earth suspension with model group,it shows that gastric lavage after poisoning can mitigate plasma concentrations of paraquat mitigation and slow down the rise in plasma concentration of paraquat;Pairwise comparisons between the 15% of bleaching earth suspension group,30% of bleaching earth suspension group and water group,The results showed that bleaching earth wash-dimensional group is better than water lavage group and 30% of bleaching earth suspension lavage are the best;Compared the plasma concentrations of paraquat after the different points of time start the gastric lavage,Shows start early group of gastric lavage(30min)was significantly lower than the late group(1h);as soon as possible after paraquat poisoning gastric lavage,gastric washing and bleaching earth concentration of 30% suspension for the best.
