Elimination Effects Of Gastric Lavage In Paraquat Lavage At Different Time Point In Swine Model

Objective:1.Establish a poisoning animai modle with paraquat intragastric administration.2.Analyze the effection on the gastric lavage at different time point.Method:1.Paraquat was infused by intragastric administration with different doses as 20mg/kg,60mg/kg,100mg/kg,establishing the paraquat poisoning modle.2.23 healthy female domestic swines were randomly divided into PQ group(n=6),PQ+lh GL group(n=6),PQ+3h GL group(n=6)and PQ+6h GL group(n=5).All the animals were administered 60mg/kg dose of 40%PQ solution through lavage.PQ+lh GL group,PQ+3h GL group and PQ+6h GL group were given gastric lavage only once with 20L warm water at 1h,3h,6h of time point after ingestion respectively.Vital signs and PQ plasma concentration were monitored continuously till 24h.LC-MS was performed to measure the plasma drug concentration.HPLC was performed to measure the washing of stomach.Result1.Without clinical manifestation of poisoning,the swines intake paraquat at dose of 30mg/kg(n=2).The swines intaking paraquat at dose of 120mg/kg(n=2)died in a short time.The survival rate of swines ingesting paraquat at dose of 60mg/kg was66.67%(4/6).2.The 24h survival rate of PQ group,PQ+6h GL group,PQ+3h GL group and PQ+lh GL group was 66.7%,83.3%,80%and 100%,respectively.The peak plasma PQ concentrations in each group were PQ(5.12±3.38)ug/ml,PQ+6h GL(3.99±1.24)?g/ml,PQ+3h GL(2.39±2.83)?g/ml and PQ+lh GL(1.96±1.83)?g/ml.The percentage of the quality of PQ in the gastric elution fluid versus the total ingestion in each group were no statistical difference;while the comparison of the content of PQ quality in the 1st half elution fluid versus in the 2nd half elution fluid in each group were:PQ+1h GL group(20.44%±5.59%)vs(4.02%±1.82%)(P<0.001),PQ+3h GL group(22.43%±3.11%)vs(3.36%±0.58%)(P<0.001),PQ+6h GL group(26.17%±17.19%)vs(7.03%±4.29%)(P<0.001)?Conclusion:1.The paraquat was given as 60mg/kg in the modle of paraquat poisoning.2.Gastric lavage during this three time point all can eliminate PQ effectively.Gastric lavage can cause significantly decline of the plasma PQ peak concentration.However,the clearance rate descended sharply at the 2nd half elution fluid in comparison with the 1st half elution fluid.
