Objective: To observe therapeutic effects of gastric lavage with Fuller’s earth combinedwith QingyiⅡ catharsis on oral paraquat poisoning in rabbits.Methods:30healthy adult Japanese white rabbits (male and female half) were randomlydivided into five experimental groups,6each group, namely group N (normal controlgroup), X group(model group), group A (Fuller’s earth gastric lavage), group B(QingyiⅡcatharsis) and group C (10%Fuller’s earth suspension liquid gastric lavage+Qingyi Ⅱ catharsis). All groups were given paraquat100mg/kg to establish the model ofacute poisoning through gavage except Group N (given saline as the same dose asexperimental groups). The group X: no gastric lavage, no catharsis. In the group A, at1hour after establishing poisoning models,30ml30%Fuller’s earth suspension liquid wasinjected into stomach, and gastric lavage with10%Fuller’s earth suspension liquid wasperformed after5minutes, and then30ml30%Fuller’s earth suspension liquid wasinjected into stomach again after gastric lavage. In the group B, at1hour after establishingpoisoning models,20ml Qingyi Ⅱ was injected into stomach through the mouth,20mleach time,2times each day, for3days. In the group C: at1hour after establishingpoisoning models,30ml30%Fuller’s earth suspension liquid was injected into stomach,and gastric lavage with10%Fuller’s earth suspension liquid was performed after5minutes,and then30ml30%Fuller’s earth suspension liquid was injected into stomach again aftergastric lavage, and Qingyi Ⅱ was injected into stomach through the mouth after10minutes,20ml each time,2times each day, for3days. Plasma paraquat concentration wasdetected with ultraviolet spectrophotometer, respectively at1h,2h,4h,8h,24h afterpoisoning in all groups. Survival was observed in rabbits, and at8days after poisoning, theanimals were euthanised by intravenous injection of air and the right lower lobe was takento make pathological section. To compare survival rates, plasma concentration change andlung tissue pathology change of lung injury in all groups. Results:1. Survival rates. Survival rates at2days after establishing poisoning models was0%in group X. In group A, the survival rates at2days after poisoning was83.3%, and50%at7days. In group B, the survival rates at2days after poisoning was33.3%, and0%at7days. However, in group C, the survival rates at2days after poisoning was100%, and83.3%at7days. The survival rates at2days after poisoning were compared among thesefour groups, group C>A group>Group B>X group (P<0.001, Fisher’sprobability method). The survival rates at7days, group C>A group>Group B (P<0.001, Fisher’s probability method), but Group B=Group X.2. Plasma concentration ofparaquat: In multiple comparison of any two groups about plasma concentration ofparaquat poisoning rabbits, group X and group A, P<0.001; group X and group B, P=0.006<0.05; group X and group C, P<0.001; group A and group B, P=0.001<0.05; group A and group C, P<0.001; group B and group C, P<0.001, suggestingcomparative differences among groups with statistical significance. The decreasedamplitude of plasma concentrations of paraquat were compared, group C>group A>Group B. The plasma paraquat concentrations of different time points were detected inthese four groups. Factorial analysis showed that P<0.001, at t2h、 t4h、t8h、t24h exceptt1h in the group A and B. There were significant therapeutic effects in the group A and Bthan the group X. A synergistic therapeutic effect was shown between Fuller’s earth gastriclavage and QingyiⅡcatharsis at t4h in the group C (P=0.034<0.05).3. Lunghistopathology: obvious expansion and hyperemia of the alveolar capillary, widenedalveolar septum, a large number of inflammatory cell infiltration, were observed in groupX and B, and there was no significant difference between the two groups. Lunghistopathology was more serious in group X and B than group C and A, and group Cimproved more obviously than group A.Conclusion: Early start of gastric lavage with Fuller’s earth combined withQingyiⅡcatharsis, can significantly improve2days and7days survival rates in oralparaquat poisoning rabbits. At the same time, it can reduce the animal’s plasmaconcentrations of paraquat, and improve lung tissue pathological changes of lung injurycompared with a single gastric lavage or catharsis. Gastric lavage with Fuller’s earthcombined with QingyiⅡcatharsis can improve the prognosis of oral paraquat poisoningrabbits synergistically.
