Experimental Study On The Effect Of Sanshen Decoction On TNF-??IL-6?IL-10 In Experimental Heart Failure Rats

Objective:By using Sanshen decoction on treating heart failure rats, to observe the level of tumor necrosis factor alpha(TNF-?), interleukin-6(IL-6),interleukin-10(IL-10) in serum and the pathological changes of hearts and lungs,and evaluate the effect of Sanshen decoction in the treatment of heart failure in rats.Methods:Adult male Wistar rats were randomly divided into 7 groups, respectively for the blank group, model group and group, enalapril group, blood circulation group,Sanshen decoction low dose group,Sanshen decoction middle dose group and Sanshen decoction high dose group.Except the blank group,others copy the model of heart failure rat by intraperitoneal injection of adriamycin solution.After the experiment began,drug intervention groups give drug lavage,model group and blank group with saline lavage for eight weeks.After that,we took blood from the abdominal aorta of rats,detected level of IL-6,IL-10,TNF-? and BNP, took the hearts and lungs of rats,and observed pathology changes of the hearts and the lungs by light microscopy after fixation and staining.Result : Serum IL-6,TNF-?,BNP,IL-10 levels in the treatment groups and model group were significantly higher than the blank group(P<0.05). Compared to other drug intervention groups,the level of IL-6,TNF-?,BNP in Sanshen decoction middle dose group,high dose group and enalapril group decreased more significantly,but IL-10 level increased more significantly(P<0.05),Sanshen decoction middle dose group,high dose group and enalapril group were no statistically significant difference between the level of serum TNF-?,IL-6,IL-10 and BNP. Then observe rats' myocardia and lungs with Light microscopy.Compared with the blank group, model group and the drug intervention groups have different damage of hearts and lungs(P<0.05),of which the damage in Sanshen decoction middle dose group,high dose group and enalapril group were lower,closed to the blank group.Conclusion : Sanshen decoction can reduce the level of heart failure rats' serum TNF-?,IL-6,increase the level of IL-10,suppress the immune inflammatory response.And Sanshen decoction can reduce the level of heart failure rats' serum BNP,improves heart failure rats' cardiac function. Sanshen decoction can improve heart failure rats' myocardial damage status and prevent myocardial fibrosis. At the same time, which also can improve the symptoms of pulmonary congestion about heart failure rats, etc.
