| Objective:To observe the influence about serum TGF-?1?MMP-9?CTGF and BNP of sanshen compound prescription on rats with chronic heart failure and the pathological changes of the rats' heart liver and lung in order to supply the theory evidence for the Chinese traditional treatment of this disease.Methods:The study include animal experiment and clinicopathologic analysis. Healthy adult rats selection Wister, the average weight 200±10g,were randomly divided into seven groups: the blank group, model group, enalapril group, promoting blood circulation group, sanshen decoction of low,medium and high dose group,each group only 12.Except the blank group, other groups were made into CHF model by injecting ADR into abdominal cavity every week. When in the first,third and sixth weeks the drug concentration are 3mg/kg,the other drug concentration are 2mg/kg. The rats were given irrigation stomach by 10ml/kg at the beginning:the blank group and model group need normal saline to be irrigated stomach;the sanshen decoction of low,medium and high dose group are respectively given by the sanshen decoction of crude drug concentration 25g/kg?50g/kg?75g/kg;promoting blood circulation group are given crude drug concentration 50g/kg.After 8 weeks,MMP-9?TGF-?1?CTGF and BNP in serum were measured by using ELISA. The heart,liver and lung were fixed and stained, then observe the change of pathology through microscope.Results:Serum BNP,CTGF,MMP-9, TGF-?1levels in the treatment group and model group were significantly higher than the blank group(P < 0.05). Compared to other drug intervention group, the levels of CTGF,MMP-9 and TGF-?1in Sanshen decoction middle dose group,high dose group and enalapril group decreased more significantly(P < 0.05). Sanshen decoction middle dose group,high dose group and enalapril group in were no statistically significant difference between the level of serum CTGF,MMP-9, TGF-?1 and BNP(p>0.05). Sanshen decoction low does group and promoting blood circulation group in were no statistically significant difference between the level of serum CTGF,MMP-9, TGF-?1 and BNP(p>0.05). Then observe rats' myocardium and lung with Ligh microscopy,compared with the blank group, model group and the drug intervention group have different damages of heart and lung(P < 0.05), of which the damage in Sanshen decoction middle dose group,high dose group and enalapril group were lower, close to the blank group.Conclusion: Sanshen decoction can reduce the level of heart failure rats' serum BNP,CTGF,MMP-9, TGF-?1 and BNP content and suppress the myocardial fibrosis and ventricular remodeling. At the same time, Sanshen decoction can reduce the damage of the heart failure rats' heart,lung and liver, which also contribute to cure heart failure rats, etc. |
